摘    要:用果蝇伴性隐性致死试验进行大蒜素的抗诱变性研究。大蒜素与甲基磺酸甲酯(MMS)同时喂饲果蝇,未能抑制MMS诱导的伴性隐性致死效应。而在先给MMS 24h后再给大蒜素则有抑制效应。大蒜素与环磷酰胺(Cy)同时给药有一定的抑制作用。提示大蒜素有一定抗诱变作用。

Abstract:The effect of garlicin on sex-linked recessive lethal (SLRL) mutations induced by direct mutagen MMS and indirect mutagen Cy in Drosophila melanogaster was studied. The adult Oregon-R male and Muller-5 virgin female Drosophila were used with 2-3-3 procedures. Garlicin 0.4 mg / ml served as negative control and 5% sucrose phosphate buffer solution as solvent control, 2 mmol/L MMS and 25 mmol/L Cy served as positjve control. The experiment was divided into 3 parts: In part 1. Drosophila were fed with 2 mmol/L MMS and 0.2 and 0.4 mg / ml garlicin for 24 h. The results showed that under experimental conditions, garlicin did not significantly inhibit lethal mutation effects induced by MMS. In part 2. After Drosophila had been fed with 2 mmol / L MMS for 24 h, they were fed with 0.2 or 0.4 mg/ml garlicin for 24 h. The results showed that garlicin reduced tptal lethality rate from 13.15% to 4.59% and 2.42% (p<0.01) respectively in the threebroods, the differences were very significant when compared with the positive control group. In part 3. Drosophila were fed with 25 mmol / L Cy and 0.2 or 0.4 mg/ml garlicin for 3 d. The results showed that garlicin reduced the total lethality rate from 3.5% to 1.99% and 1.42% respectively in the three broods, the differences had no significance compared with the positive control group. However, the frequency inhibition of about 50% showed some effects of garlicin. In view of the above observations, it might be concluded that garlicin is a bioantimutagen; the possible mechanisms may be due to the stabilization of DNA, the improvement of DNA fidelity of error-free repair function and the suppression of error-prone repair function.
