甘草提取物对胺碘酮在大鼠体内药动学的影响

摘    要:目的研究甘草对胺碘酮在大鼠体内药动学的影响。方法 14只实验大鼠随机分为对照组与实验组,分别予0.9%氯化钠溶液和甘草提取物(0.5g·kg^-1,qd×7d)后,胺碘酮灌胃给药按时间点连续采样,采用HPLC法测定血药浓度。计算并比较主要药动学参数。结果对照组和实验组的胺碘酮主要药动学参数ρmax、tmax、t1/2、AUC0→24h2、AUC0→∞、CL/F和MRT差异均无统计学意义(P〉0.05)。结论甘草连续给药7d后不影响胺碘酮在大鼠体内的药动学特征。

Abstract:AIM To study the effect of licorice on amiodarone pharmacokinetics in rats.METHODS Experimental rats were randomly divided into the control group and test group,that were administrated respectively with normal saline and extract of licorice.After pretreated with extract of licorice(0.5 g·kg^-1,qd) for 7 d,amiodarone was given to the rats by intragastric administration.The plasma concentrations of amiodarone were determined by HPLC.Pharmacokinetic parameters between 2 groups were compared.RESULTS There was no significant difference between 2 groups in the main pharmacokinetic parameters of amiodarone,such asρ_(max),t_(max),t_(1/2),AUC_(0→24h),AUC_(0→∞),CL/F and MRT (P0.05).CONCLUSION The results suggest that treatment with extract of licorice(0.5 g·kg~(-1),qd) for 7 d does not affect the pharmacokinetics of amiodarone in rats.
