鹌鹑流感病毒SA受体的分布特点及其在流感病毒生态系统中的作用

摘    要:目的探讨鹌鹑流感病毒受体分布特点及其在流感病毒生态系统中的位置和作用。方法应用亲和组化法检测鹌鹑呼吸道和消化道流感病毒SA受体的分布,用荧光素Alexa488标记的禽流感病毒H9N1、人流感病毒H1N1分别与鹌鹑呼吸道、消化道各解剖部位结合。结果鹌鹑呼吸道均表达SAα-2,3Gal受体和SAα-2,6Gal受体,SAα-2,3Gal受体在呼吸道气管、支气管、次级支气管和消化道结肠上皮细胞均呈弱阳性分布,而副支气管呈阳性弥漫分布;SAα-2,6Gal受体在呼吸道气管、支气管和次级支气管多呈强阳性弥漫分布,而在副支气管、消化道结肠均为缺乏或仅极少量分布。禽流感病毒H9N1、人流感病毒H1N1均可与呼吸道和消化道上皮细胞结合。结论鹌鹑同时表达SAα-2,3Gal受体和α-2,6Gal受体,能与禽流感病毒和人流感病毒结合,有助于病毒基因重配,可充当产生人类流感大流行潜在的中间宿主。

Abstract:Objective To study the distribution characteristics of influenza virus receptor in quail,and the position and role of quail in influenza virus ecosystem.Methods The distribution of sialic acids receptor in quail respiratory tract and alimentary tract was investigated by using lectin histochemical staining.The epithelial cells in respiratory tract and alimentary tract of quail were bind by Alexa 488 labelled avian influenza H9N2 virus and human influenza H1N1 virus,respectively.Results Both SAα-2,3Gal receptor and SAα-2,6Gal receptor were expressed in quail respiratory tract.The SAα-2,3Gal receptor was detected in a few of the trachea,bronchus,second bronchus and colon,but was prevalent in parabrochus of quail.The SAα-2,6Gal receptor was prevalent in the trachea,bronchus and second bronchus,but was deficient or infrequent in the parabrochus and colon.Furthermore,Alexa 488 labelled avian influenza H9N2 virus and human influenza H1N1 virus both could bind to epithelial cells in respiratory tract and alimentary tract of quail.Conclusion These results suggested that quail,which carry both SAα-2,3Gal and SAα-2,6Gal receptor compatible with binding of avian and human influenza viruses,could provide an environment of reassortants between avian and human influenza viruses and act as an intermediate host to generate influenza virus with pandemic potential.
