四逆汤对肾性高血压大鼠肾及心脑病变的影响

摘    要:目的探讨四逆汤对高血压大鼠肾及心脑等器官病理学改变的影响。方法取健康雌性Wistar大鼠24只,随机分为正常对照组、肾性高血压组、四逆汤治疗组,每组8只。应用双肾动脉夹闭法建立肾性高血压动物模型,应用RBF-2型大鼠尾动脉血压计测量血压。四逆汤治疗组给予四逆汤灌胃4周,肾性高血压组同期给予生理盐水灌胃,正常对照组不做任何处理。观察各组大鼠肾及心脑的组织病理学改变。结果四逆汤治疗组血压降至正常;肾性高血压组大鼠肾、心、脑结构较正常对照组均有不同程度的改变;四逆汤治疗组大鼠肾脏入球小动脉壁厚度较肾性高血压组明显变薄(F=11.086,q=2.82,P〈0.05),心肌细胞横截面积显著减小(F=13.607,q=2.83,P〈0.05),脑血管病理改变较对照组无显著性差异。结论四逆汤对高血压具有一定的治疗作用,在降压的同时能够一定程度减轻肾、心、脑的病理改变。

Abstract:ObjectiveTo study the effects of Sinitang on histopathologic changes in kidney,heart and brain in renal hypertensive rat.MethodsHealthy female Wistar rats(n=24) were evenly and randomly divided into normal control(NC) group,renal hypertension(RH) group and Sinitang-treated(ST) group.The renal hypertensive models were established by double-cliping the renal arteries.Sinitang was given through an intragastric tube to the rats in ST group for four weeks,while normal saline was given to those in RH group at the same time,and no any treatment given to the rats in NC group.The histopathologic changes of each group were then observed.ResultsExtensive pathological changes were noted in the kidney,heart and brain in RH group compared with that in NC group.The pathologic structure changes of kidney in ST group were obviously lightened than that in RH group,and the wall of afferent arteriole of glomerulus of ST group was thinner than that in RH group(F=11.086,q=2.82,P<0.05);the average cross section area of myocardial fiber in ST group was smaller than that in RT group(F=13.607,q=2.83,P<0.05);while no significant difference in brain histopathologic changes was noticed.ConclusionSinitang can relieve the pathologic changes in kidney and heart of renal hypertensive rats by reducing blood pressure.
