糖基化终产物对兔股动脉内皮依赖性血管舒张功能的影响

摘    要:目的 研究不同浓度的糖基化终产物(AGEs)对兔股动脉内皮依赖及非内皮依赖的血管舒张功能的影响。方法 采用家兔股动脉恒流灌注模型,在兔股动脉远心端连接压力换能器,测定兔股动脉灌注压;从而间接反映兔股动脉远心端血管舒张功能。记录AGEs阻断前后不同浓度的乙酰胆碱(Ach)及不同浓度的硝普钠(SNP)灌注兔股动脉的压力曲线,观察AGEs对不同浓度Ach所引起的内皮依赖性血管舒张及SNP引起的内皮非依赖性血管舒张的影响,以及L-精氨酸(L-arginine)对这种影响的作用。结果 Ach及SNP均引起浓度依赖的血管舒张,AGEs抑制Ach产生的血管舒张作用并呈浓度依赖性,L-arginine不能改善AGEs对Ach引起的血管舒张的抑制作用。AGEs对SNP引起的血管舒张作用没有影响。结论AGEs通过减少NO的合成抑制内皮依赖性血管舒张,NO前体物L-arginine对AGEs的作用无明显影响。

Abstract:Objective To determine whether AGEs inhibits endothelium - dependent vasorelaxation of rabbit femoral arteries and it' s possible mechanisms. Methods Constant perfusion model of rabbit femoral artery was used. The effects of AGEs on endothelium - dependent vasorelaxation induced by different concentration of acetylcholine (Ach) and endothelium - independent vasorelaxation induced by different concentration of sodium nitroprusside (SNP) were observed. In addition, effects of L-arginine on the inhibition of Ach - induced vasorelaxation by AGEs also were observed. Results Ach induces concentration -dependent vasorelaxation, AGEs significantly inhibits this sort of vasorelaxation by Ach, and this inhibition effect correlates with AGEs concentration. L - arginine can not prevent this inhibition. SNP also induces concentration - dependent vasorelaxation, but AGEs has no effect on vasorelaxation induced by SNP. Conclusion AGEs inhibits endothelium - dependent vasorelaxation of rabbit femoral arteries through reducing endogenous NO synthesis.
