摘    要:目的 检测S期激酶相关蛋白2(Skp2)和抑癌基凶p27kip1在肾细胞癌(RCC)中的表达,探讨其与肾细胞癌生物学特征的关系及意义.方法 采用组织芯片技术及免疫组化SP法检测Skp2和p27kip1在80例肾细胞癌组织和40例癌旁正常肾组织中的表达,并做统计学分析.结果 (1)Skp2在肾细胞癌中的表达率高于正常肾组织(P=0.025);随肿瘤恶性程度增加,Skp2阳性表达率升高(P=0.002),其表达率与肾癌患者的年龄、性别、临床分期、淋巴结转移及肿瘤大小无关(P>0.05).(2)p27kpi1在肾细胞癌中的表达率低于正常肾组织(P=0.007);p27kip1阳性表达率与肿瘤分级及临床分期呈负相关((P<0.05),与肾癌患者的年龄、性别、淋巴结转移及肿瘤大小无关(P>0.05).(3)Skp2与p27kip1表达呈显著负相关(r=-0.273,P=0.014).结论 肾细胞癌中Skp2蛋白的过度表达与p27kip1蛋白降解增强有关,提示Skp2可能在肾细胞癌发生中扮演着重要角色.

Abstract:OBJECTIVE: To detect the expression of skp2 and p27kip1 in human renal cell carcinoma (RCC) using tissue chip technique, and evaluate the relationship between the proteins and the biological behavior of RCC. METHODS: Tissue chip technique and immunohistochemical SP method was used to detect the expression of skp2 and p27kip1 in normal and tumor tissues. RESULTS: The positivity rate of Skp2 in RCC was significantly higher than that in normal renal tissues (P=0.025). The positivity rate of Skp2 expression in RCC was significantly correlated to poor differentiation of the tumor (P=0.002), and was not associated with the patients gender, age, tumor size, lymph node metastasis and stages of RCC (P>0.05). The positivity rate of p27kip1 in RCC was significantly lower than that in normal renal tissues (P=0.007). The positivity rate of p27kip1 expression was inversely correlated to the malignancy and stage of RCC (P<0.05), but not with the patients' age, gender, lymph node metastasis and tumor size (P>0.05). An inverse correlation was noted between Skp2 and p27kip1 expressions (r= -0.273, P=0.014). CONCLUSION: Overexpression of Skp2 protein may lead to decreased p27kip1 level in RCC, indicating its involvement in the carcinogenesis and development of RCC.
