黄芪桂枝五物汤止痒作用的实验研究

摘    要:目的研究黄芪桂枝五物汤的止痒作用及其机制。方法采用尾静脉注射低分子右旋糖酐诱发小鼠阵发性皮肤瘙痒,记录小鼠30min内的阵发性皮肤瘙痒发作次数及时间,并测定小鼠血中组胺、TXB2、6-keto-PGF1α的含量。结果与模型组相比,黄芪桂枝五物汤各剂量组均能明显抑制阵发性皮肤瘙痒发作次数,减少持续时间,差异有统计学意义(P0.01);黄芪桂枝五物汤各剂量组均能降低血清中组胺含量(P0.05或P0.01),但以中剂量组最佳;黄芪桂枝五物汤各剂量组TXB2均显著降低(P0.01);而黄芪桂枝五物汤大、中剂量组6-Keto-PGF1α显著升高(P0.05或P0.01),小剂量组6-Keto-PGF1α含量变化无统计学差异(P0.05)。与苯海拉明组相比,在瘙痒发作次数和持续时间上,黄芪桂枝五物汤中剂量组(P0.05)较高低剂量组(P0.01)差异小。结论黄芪桂枝五物汤有明显的止痒作用,其机理可能与拮抗组胺释放和促进血循环均有关系,但与抑制组胺释放关系最为密切;其止痒作用与剂量有一定的相关性。

Abstract:Objective To study the antipruritic effect and its mechanism of huangqi guizhi wuwu decoction.MethodsAdopt the injection of low molecular dextran in the tails to induce paroxysmal skin pruritus in mice,record the attack frequency and time of paroxysmal skin pruritus in 30 min and detect the contents of histamine,TXB2 and 6-keto-PGF1α in blood of mice.ResultsCompared with model group,huangqi guizhi wuwu decoction groups of any dose inhibited apparently the attack frequency of paroxysmal skin pruritus and reduced the persistent time,indicating statistical significant difference(P0.01).Additionally,it decreased histamine content in serum(P0.05 or P0.01),of which,the results in the middle dose group were the best.TXB2 level decreased significantly in all the dose groups(P0.01),6-keto-PGF1α increased significantly in the high and middle dose groups(P0.05 or P0.01),and there was no statistical difference in 6-keto-PGF1α level in low dose group(P0.05).Compared with diphenhydramine group,the differences in pruritus frequency and persistent time of pruritus were less in middle dose group(P0.05)rather than in high and low dose groups(P0.01).ConclusionHuangqi guizhi wuwu decoction has obvious antipruritic effect,which is probably relevant with the antagonism of histamine release and the promotion of blood circulation.But,its inhibition on histamine release is mostly involved in the mechanism.Additionally,a certain correlation appears between its antipruritic effect and the dose of drug.
