宫颈癌组织中FBW7和c-Myc的表达及相关性分析

摘    要:目的:检测F盒/WD40重复域蛋白7(FBW7)和c-Myc在宫颈癌组织中的表达情况及临床意义,并分析两者表达的相关性。方法:收集我院2008年1月~2012年12月病理科存档的宫颈癌和正常宫颈组织标本70例,采用免疫组化SP法分别检测46例宫颈癌及24例正常宫颈组织中FBW7及cMyc的表达情况。结果:FBW7和c-Myc在宫颈癌组织中的阳性表达率分别为30.4%(14/46)和69.6%(32/46),在正常宫颈组织中的阳性表达率分别为66.7%(16/24)和25.0%(6/24),两组比较差异有统计学意义(χ2=8.454,P=0.004;χ2=12.622,P=0.000)。FBW7表达与宫颈癌临床分期(P=0.036)和病理学分级(P=0.023)显著相关,c-Myc表达与宫颈癌临床分期(P=0.036)、淋巴结转移(P=0.003)和病理学分级(P=0.003)也显著相关。宫颈癌组织中FBW7与c-Myc蛋白表达呈显著负相关(r=-0.667,χ2=6.694,P=0.010)。结论:宫颈癌组织中FBW7低表达,且与c-Myc表达呈负相关,提示FBW7可能通过调控c-Myc泛素化降解参与肿瘤发生、发展。

Abstract:Objective:To investigate the expression of FBW7and c-Myc and their clinical significance and correlation analysis in cervical cancer tissues.Methods:FBW7and c-Myc expression was detected in 46samples of surgically resected cervical cancer and 24samples of normal cervix tissues using immunohistochemical assay.Results:The positive rate of FBW7and c-Myc in cervical cancer tissues(30.4% and 69.6%)were significantly different from those in the normal cervix tissues(66.7%and 25.0%)(χ2= 8.454,P=0.004;χ2=12.622,P=0.000).The expression of FBW7was significantly correlated with clinical stages(P=0.036)and pathological grades(P=0.023),and the expression of c-Myc was significantly correlated with clinical stages(P=0.036),lymph node metastasis(P=0.003)and pathological grades(P =0.003).FBW was negatively correlated with c-Myc protein expression in cervical cancer tissues.Conclusion:The low-expression of FBW7is negatively correlated with c-Myc and is associated with poor clinical parameters in cervical cancer.FBW7may promote cancer progression by regulating c-Myc ubiquitination degradation in cervical cancer.
