摘    要:【目的】正交实验法优选肉豆蔻中安五脂素的提取工艺。【方法】建立肉豆蔻中安五脂素的HPLC含量测定方法,监测提取工艺中安五脂素的含量,采用L9(34)正交试验设计法,考察乙醇浓度、固液比、提取时间、提取次数对肉豆蔻中安五脂素提取的影响。【结果】安五脂素含量测定方法为标准曲线回归方程:Y=2×107X-57655(R2=0.9996),安五脂素在0.791~3.164μg范围内线性关系良好,安五脂素的平均回收率为100.9%。通过正交实验法确定肉豆蔻中安五脂素的最佳提取工艺为12倍量的95%的乙醇回流提取60 min,提取次数为2次。【结论】该提取工艺简便易行,为肉豆蔻药材的综合利用提供了依据。

Abstract:Objective] To establish the optimum extraction process of anwulignan in Myristica fragrans Houtt. by orthogonal experimental method. Method] HPLC was used to determine the content of anwulignan and orthogonal test was used to optimize the extraction technology, concentration of ethanol, the ratio of material to liquid, extraction duration and extraction times on extraction rate of anwulignan was studied. Result] The standard curve of anwulignan regression equation: Y=2 x 107X-57655(R2=0.9996), the anwulignan in the range 0.791-3.164 μg a good linear relationship, the average recovery of anwulignan was 100.9%. The optimum extraction processes were as follows: the ratio of solid to liquid was 1:12, concentration of ethanol is 95%, the extraction time is 60 mins and extraction for two times. Conclusion ] The extraction process is simple and easy industrialization, laying the foundations for the comprehensive utilization of Myristica fragrans Houtt.
