MMP20在鼻咽癌中表达及与预后关联性分析

目的 探讨MMP20在鼻咽癌中的表达以及与临床参数、预后之间的关系。方法 收集南方医科大学南方医院病理科共127例鼻咽癌组织和37例正常鼻咽组织,利用免疫组化检测MMP20蛋白表达。采用卡方和Kaplan Meier生存曲线统计分析MMP20蛋白表达变化以及鼻咽癌患者临床参数、预后之间的关系。稳定沉默鼻咽癌细胞MMP20的表达,比较单克隆形成能力。结果 免疫组化分析检测显示,MMP20蛋白在鼻咽癌与正常鼻咽组织的细胞浆中均表达,鼻咽癌组织中的表达明显高于鼻咽组织。MMP20的高表达与鼻咽癌患者的T、N和TNM分期呈明显正相关。MMP20蛋白升高明显缩短鼻咽癌患者的生存时间,显示MMP20表达是预测鼻咽癌患者预后的预测因子。沉默MMP20表达会导致鼻咽癌细胞增殖能力下降。结论 MMP20蛋白在鼻咽癌中高表达并导致了患者预后不良。

Correlation analysis of MMP20 expression and prognosis in nasopharyngeal carcinoma

Objective To mainly explore the expression of MMP20, clinical significance and prognosis in nasopharyngeal carcinoma (NPC). Methods Totally 127 paraffin-embedding endometrial carcinoma tissues and 37 paraffin-embedding nasopharynx tissues (NP) were collected from Southern Medical University Nanfang Hospital. MMP20 protein expression was tested in these tissues. Chi-Square and Kaplan Meier survival curve were used to analyze the expression change and its correlation with clinical pathology features and prognosis in NPC. Growth of NPC cells were compared between groups with or without MMP20 stably knocked down by colony-forming assay. Results Immunohistochemistry analysis showed that MMP20 protein was stained in cytoplasm in both NPC and NP tissues. Statistical analysis showed that MMP20 was upregulated in NPC tissues compared to NP tissues. Furthermore, elevated MMP20 protein expression was positively associated with T and N classification as well as TNM stage in NPC. In addition, upregulated MMP20 protein expression markedly decreased the survival time of NPC patients, multivariate analysis showed that MMP20 expression was a potential independent predictor of prognosis in NPC patients. Stably knocking down the expression of MMP20 decreased the growth of NPC cells. Conclusion Highly expressed MMP20 was a potential poor prognostic factor in NPC .
