人端粒保护蛋白1与冠状动脉病变的关系

摘    要:目的 探讨人端粒保护蛋白1(hPOT1)的表达水平与冠状动脉粥样硬化病变发生的关系。方法 选取山东大学第二医院心脏内科住院患者79例,通过冠状动脉造影术,分为冠心病组(n=49)和对照组(n=30),分离外周血白细胞,应用实时荧光定量PCR(qRT-PCR)法检测hPOT1 mRNA表达水平,研究对象均测定身高、体质量、血压等指标,检测空腹血糖、血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)等指标。结果 冠心病组hPOT1 mRNA表达量与对照组相比明显减低(P<0.05);hPOT1 mRNA表达水平与冠心病的易患因素高血糖(r=-0.264,P<0.05)、高胆固醇(r=-0.213,P<0.05)及高血压(r=-0.331,P<0.01)分别呈负相关;多因素Logistic回归分析显示,hPOT1为冠心病发生的保护性因素(OR=0.871, 95%CI=0.852~0.900,P<0.05 )。结论 hPOT1表达水平减低与冠状动脉粥样硬化病变的发生及其易患因素密切相关。

Abstract:Objective To explore the relationship between the expression of human protection of telomeres 1 (hPOT1) and pathogenesis of coronary atherosclerosis. Methods A total of 79 in-patients in the Department of Cardiology of the Second Hospital of Shandong University were recruited. After coronary angiography, 49 patients were taken in the coronary atherosclerosis group, while the other 30 patients were taken as the control group. The height, weight, blood pressure, blood sugar, total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) of all cases were evaluated. Expression of hPOT1 mRNA was determined by real-time quantitative PCR (qRT-PCR) method. Results The level of the hPOT1 mRNA in the coronary atherosclerosis group reduced significantly (P<0.05); the expression of hPOT1 mRNA were negatively correlated with hyperglycemia (r=-0.264, P<0.05), high TC (r=-0.213, P<0.05) and hypertension (r=-0.331, P<0.01). Logistic regression analysis showed that hPOT1 was a protective factor for coronary atherosclerosis, (OR=0.871, 95%CI=0.852-0.900, P<0.05). Conclusion The low expression of hPOT1 is positively correlated with the pathogenesis and risk factors of coronary atherosclerosis.
