家兔虹膜色素上皮细胞移植的实验研究

摘    要:目的比较直接分离和体外培养的继二代有色素家兔虹膜色素上皮(IPE)细胞移植于无色素家兔视网膜下间隙后的存活状况。方法分别选用经酶-显微解剖-酶分离法直接分离的有色素家兔虹膜色素上皮细胞,和经过体外培养至继二代的有色素家兔虹膜色素上皮细胞,制成相同密度的IPE细胞悬液。采用术中检眼镜定位的外路移植法将其植入无色素家兔视网膜下间隙。术后每日直接检眼镜观察眼底,并分别于术后的第1、2、4、8、12、16周对兔眼移植区作光镜检查。结果两种不同方式制备的有色素家兔虹膜色素上皮细胞均可在无色素家兔视网膜下间隙存活。其中,经体外培养的虹膜色素上皮细胞移植后呈较均匀的单层排列。植入的虹膜色素上皮细胞在观察期内均未见明显炎症和排斥反应。结论检眼镜定位的外路法可以将供体家兔IPE细胞成功移植在受体眼视网膜下间隙,经体外培养的继二代家兔虹膜色素上皮细胞存活形态好于直接分离的家兔虹膜色素上皮细胞。

Abstract:Objective To compare the survival situation between the direct isolation and after cell transplantation of IPE of secondary generation in vitro culture. Methods The IPE were isolated with enzyme-anatomy-enzyme method and cell suspension was prepared with IPE after secondary generation in vitro culture. The cell transplantation was performed to the rabbit without pigment at inferior gap of retina with outside approach method. The eyeground was detected directly and with light microscope detection with transplantation area at 1, 2, 4, 8, 12, and 16 weeks.Results The IPE with different preparations could be survived at inferior retina. The IPE after vitro culture could be homogenous and single layer. The transplanted IPE had no reaction of inflammation and rejection. Conclusion The outside approach with ophthalmoscope might transplant the IPE to inferior retina. The survival situation of transplantation of secondary generation of vitro culture might be better than direct isolation.
