银杏提取物(GBE)对人胶质瘤U251细胞增殖凋亡的影响

目的观察利用银杏提取物对人脑胶质瘤U251细胞增殖与凋亡的影响。方法将传代的U251细胞随机分为四组,A组加培养液,B、C、D组分别加浓度为50mg/ml、100mg/ml、200mg/ml的银杏叶提取物;观察倒置显微镜下TrypanBlue染色后各组U251脑胶质瘤细胞形态,用MTT法测算U251细胞增殖率,用流式细胞仪检测U251细胞凋亡率。结果 B、C、D组细胞增殖指数逐渐下降,与A组比较,P均<0.05;A、B、C、D组细胞凋亡率分别为0.3%±0.3%、10.6%±1.7%、21.9%±4.3%、33.5%±2.1%。B、C、D组与A组比较,P均<0.05。结论银杏叶(GBE)可抑制人脑胶质瘤U251细胞增殖,并诱导其凋亡。

Research on influence of Ginkgo Biloba Extracts on proliferation and apoptosis of human glioblastoma U251 cells

Objective ] To observe the effectof proliferation and apoptosis on U251 glioma by Ginkgo Biloba Extracts. Methods ] Divide the handed U251 cells into four groups randomly. Add nutrientsolution to A,while add activated GBE（50mg/ml）0.5ml to B, GBE（100mg/ml）O.Sml to C,and activated GBE（200mg/ml）0.Sml to D. To observe the form of cells of U251 glioma cells under the microscope. Calculate the proliferation rate of U251 by MTYand analysis the apoptosis of 1251 cells by the flow cytometric. Resluts ] The index of cell proliferation of B, C, D goroup declined Gradually. Compared B, C,, D with A, P 〈 0.05.The cell apoptosis rate of group A, B, C,, D was0.3% ±0.3%, 10.6% ± 1.7%, 21.9%± 4.3%, 33.5% ± 2.1% respectively.Compared B, C,, D with A, P 〈 0.05. Conclusion ] Ginkgo Biloba Extracts can restrain the proliferation of U251 glioma and also lead to the apoptosis of U251 cells.
