摘    要: 目的 通过建立裸鼠皮下肿瘤生长模型,研究吲哚胺2,3-双加氧酶(IDO)在裸鼠体内对人宫颈癌细胞的生长促进作用。方法 IDO基因表达质粒和空白质粒分别稳定转染至人宫颈癌SiHa细胞。应用Western blot检测IDO在SiHa、SiHa/Mock 和SiHa/IDO细胞中的表达情况,用MTT试剂盒检测3种肿瘤细胞体外生长速度。种植3种肿瘤细胞至裸鼠皮下,比较其在裸鼠体内的生长速度。结果 IDO蛋白在SiHa/IDO细胞中表达阳性,在SiHa和SiHa/Mock细胞中无表达。3种肿瘤细胞体外生长曲线无统计学差异(P>0.05)。3周后接种SiHa/IDO细胞的肿瘤与接种SiHa和SiHa/Mock细胞的肿瘤相比生长迅速,差异有统计学意义(P<0.05)。结论 IDO可作为宫颈癌评价预后的指标之一,也可能是宫颈癌基因治疗潜在的靶点。

Abstract:Objective To investigate the effects of indoleamine 2,3-dioxygenase(IDO) expression on growth of human cervical cancer xenograft in nude mice.Methods SiHa cells were stable transfected with IDO gene or Mock vector.Western blot analysis was used to detect IDO expression in SiHa,SiHa/Mock and SiHa/IDO cells.Cell growth curves were performed using MTT assay.Subcutaneous tumor growth curve were performed according to the xenograft tumor volumes of the three groups.Results IDO was only detected in the cells transfected IDO expression vector(SiHa/IDO).There was no intergroup difference in doubling time in vitro(P > 0.05).The subcutaneous tumor volume of SiHa/IDO group was significantly larger than control(SiHa and SiHa/Mock) group(P < 0.05).Conclusion IDO acts not only as a prognostic factor but also as a potential molecular therapeutic gene target that against cervical cancer.
