微碱法提取山药多糖复合物的工艺研究

摘    要:目的:研究采用微碱法进一步提取山药多糖复合物的最佳工艺条件。方法:以多糖提取率为指标,采用单因素试验和四因素三水平正交试验,研究提取时间(h),提取温度(℃),料液比(g/mL)和提取pH值对多糖复合物提取率的影响。结果:微碱法提取山药多糖复合物的最优工艺条件为:提取温度为50℃,提取时间为1.5 h,提取料液比(g/mL)为1∶15,pH值为10,此时山药多糖复合物最高提取率为5.739%。结论:优选的提取条件稳定可行。

Abstract:Objective :To optimize the extraction conditions of polysaccharide compound from yam under mild alkaline condition. Methods:Single factor test and orthogonal design test of L9(34)were adopted, and extraction rate of polysaeeharide was taken as index.Effects of extraction time,extraction temperature, solid-liquid ratio and pH value on extraction rate of polysaccharide were observed. Results :The optimal conditions for yam by orthogonal test were achieved as follows: extracting for 90 min with extraction temperature at 50 ~C, solid-liquid ratio was at 1:15(g/mL), pH value was at 10.Under above conditions,the extraction rate of yam polysaccharide was 5.739%. Conclusion:The optimized extraction conditions were stable and feasible.
