摘    要:为利用三维重建工具软件对成年SD大鼠部分脑干、中脑导水管及红核结构进行三维重建,并对重建的红核进行观察和测量,探索了一种方法,即:制作大鼠脑干连续冰冻切片,Nissl染色后进行分步显微摄像并进行拼接;扫描大鼠脑定位图谱相关红核切面图像;利用PhotoshopCS3软件将上述获得的连续图像分别进行图像后处理,并利用3D-DOCTOR4.0软件分别对上述连续切片图像进行配准、分割和三维重建,对两种不同方式重建的红核进行比较。结果显示,利用大鼠脑干连续冰冻切片和大鼠脑定位图谱可以对SD大鼠红核等结构进行三维重建,重建出的结构具有立体感。可以在PC机中自由观察和测量,两种重建方法得到的红核形状大小基本类似。因此,得出结论:利用大鼠脑干的连续冰冻染色切片可以对大鼠红核进行三维重建。

Three dimensional reconstruction of red nucleus in normal adult Sprague-Dawley rat based on serial sections

Abstract:Objective To make three dimensional (3D) reconstruction of the brain stem, central aqueduct and red nucleus of normal adult SD rat, and to observe and measure the reconstructed red nucleus in personal computer (PC). Methods The serial cryosections of rat' s brain stem were prepared, and the micro-photographs of which were taken by steps and then stitched after Nissl staining. Meanwhile, the red nucleus images were scanned from the Rat Brain in Stereotaxic Coordinates. PhotoshopCS3 software was used to process the above images, and 3D-DOCTOR 4.0 software was used to make registration, segmentation and reconstruction. The reconstructed red nucleus form both above mentioned approaches were compared. Results The reconstructed red nucleus based on serial cryosections of rat' s brain stem and scanned images from atlas were solid and could be observed and measured arbitrarily in PC, with similar shape and size. Conclusion The rat' s red nucleus can be reconstructed based on serial cryosections of rat' s stained brain stem.
