小蓟酸水提取物对巴马小型猪心肌的影响

摘    要:目的研究小蓟酸水提取物对巴马小型猪心肌的影响。方法健康巴马小型猪30只随机分为正常对照组、异丙肾上腺素阳性对照组(阳性对照组)和小蓟酸水提取物(高、中、低剂量)干预组(高、中、低剂量组),每组各6只。分别予以生理盐水、异丙肾上腺素(1.0 mg/kg)、小蓟酸水提取物高(2.0 mL/kg)、中(1.0 mL/kg)、低(0.5 mL/kg)剂量腹腔注射,1次/d,连续给药7 d后,检测其心电图、血清心肌酶谱和心肌氯化三苯基四氮唑(TTC)染色结果。结果与正常对照组比较,低剂量组的心电图与正常对照组基本一致,中剂量组的心电图S-T段稍有抬高,高剂量组心电图呈典型心肌缺血。血清心肌酶各指标,低剂量组各指标与正常对照组比较,差异均无统计学意义(P〉0.05);中剂量组乳酸脱氢酶(LDH)、肌酸激酶同工酶(CK-MB)明显高于正常对照组,高剂量组LDH、CK-MB、肌酸激酶(CK)、天门冬氨酸氨基转移酶(AST)明显高于正常对照组,差异均有统计学意义(P〈0.05)。心肌TTC染色结果表明中、高剂量组都有因缺血而坏死的心肌。结论通过注射小蓟酸水提取物能在一定程度上诱发巴马小型猪产生心肌缺血,缺血程度与剂量有一定关系。

Abstract:Objective To study the effects of herba cirsii extracts with acid water on myocardium in Bama miniature pigs. Methods 30 Healthy Bama miniature pigs were divided into five groups randomly, normal control group, isoprenaline control group(positive control group), high dose, medium dose, low dose herba cirsii extracts with acid water group(high dose, medium dose, low dose group), with 6 pigs in each group. They were given to corresponding drugs normal saline, isoprenaline(1.0 mg/kg), high dose(2.0 mL/kg), medium dose(1.0 mL/kg), low dose(0.5 mL/kg) herba cirsii extracts with acid water] by intraperitoneal injection once a day. After 7 days, the changes of electrocardiogram, serum myocardial enzymes and triphenyltetrazolium chloride(TTC) dyeing of myocardium were observed. Results Compared with normal control group, the electrocardiogram of low dose group was as same as normal control group, but the S-T segment of electrocardiogram in medium dose group raised slightly and the electrocardiogram in high dose group showed typical myocardial ischemia. The indexes of serum myocardial enzymes, all indexes of low dose group showed no statistically significant difference compared with normal control group(P〉0.05), but the figures of lactate dehydrogenase(LDH) and creatine kinase isoenzyme(CK-MB) in medium dose group were higher than these of normal control group, and the figures of LDH, CK-MB, creatine kinase(CK) and aspartate amino transferase(AST) in high dose group were were higher than those of normal control group, the differences were statistically significant(P〈0.05). In addition, some myocardium in medium and high dose groups were examined ischemia by TTC dyeing. Conclusion Herba cirsii extracts of acid water can induce myocardial ischemia in Bama miniature pigs in a certain extent, and there is certain correlation between the degree of ischemia and dose.
