丙烯酰胺对小鼠海马神经胶质酸性蛋白表达的影响

摘    要:目的研究丙烯酰胺对小鼠海马神经胶质酸性蛋白(GFAP)表达的影响。方法健康雄性昆明小鼠40只,随机分为对照组和5 mg.kg-1.d-1、30 mg.kg-1.d-1、60 mg.kg-1.d-13个剂量实验组,每组10只。对照组小鼠腹腔注射生理盐水;实验组小鼠分别代以5 mg.kg-1.d-1、30 mg.kg-1.d-1、60 mg.kg-1.d-1丙烯酰胺水溶液腹腔注射。5 d时分别处死各组小鼠,取出大脑,Zambon i固定液固定,常规冰冻制片,免疫组织化学染色,光镜观察。结果5 mg.kg-1.d-1、30 mg.kg-1.d-1、60 mg.kg-1.d-13个剂量实验组小鼠海马中GFAP阳性表达比对照组增强(P<0.05),但5 mg.kg-1.d-1、30 mg.kg-1.d-1剂量组间未见明显差异(P>0.05);60 mg.kg-1.d-1实验组强于5 mg.kg-1.d-1、30 mg.kg-1.d-1剂量组(P<0.05)。结论丙烯酰胺染毒后小鼠海马神经胶质酸性蛋白表达增高,提示其对小鼠海马有一定的损伤作用。

Abstract:Objective To study the influences of Acrylamide on the expression of Glial Fibrillar Acidic Protein (GFAP) in murine Hippocampus. Methods Forty healthy male mice of Kunming Genus werw randomly divided into four groups : one control group and three study groups, ten mice in each group. The control group were injected peritoneal physiological saline injection. Three study groups were injected acrylamide solution 5 mg·kg^-1·d^-1,30 mg·kg^-1·d^-1 ,and 60 mg·kg^-1·d^-1, respectively. On the 5th day,brains of each group were taken out quickly,fixed by Zamboni fixation solution, made into conventional freezing moviemaking, stained by immunohistochemistry staining, and then observed by light microscope. Results The positive expressions of GFAP in the three study groups were more increased than that of the control group( P 〈 0.05 ) ,but there were no significant difference among themselves(P 〉0.05 ). The 60 mg·kg^-1·d^-1 study group was better than the other two study groups( P 〈 0.05 ). Conclusion If mouse are injected Acrylamide solution, GFAP expression will be increased. Therefore, it hints that Acrylamide solution injection has a limited impairment effect on routine hippocampus.
