黄芪多糖对糖尿病大鼠肺组织中肾素-血管紧张素系统活性的影响

摘    要:目的探讨黄芪多糖对糖尿病大鼠肺组织中肾素-血管紧张素系统活性的影响。方法取4周龄健康SD大鼠,分为正常对照组、糖尿病模型组、培哚普利组、黄芪多糖组。分别给予胃管灌注生理盐水(正常对照组、模型组)、培哚普利2mg/kg(培哚普利组)、黄芪多糖1g/kg(黄芪多糖组),1次/d,每周灌胃5次。8周后处死大鼠,留取肺组织进行组织匀浆,测定血管紧张素Ⅱ(Angiotensin Ⅱ,AngⅡ)浓度,通过苏木精-伊红染色观察肺组织病理变化,免疫组化和图像分析法检测肺组织中AngⅡ的Ⅰ型受体(Angiotensin Ⅱ receptor Ⅰ,ATIR)的表达。结果①糖尿病模型组肺组织肺泡萎陷、破坏,肺泡间隔增宽,培哚普利组、黄芪多糖组大鼠肺泡结构改善。②AngⅡ浓度:模型组、培哚普利组、黄芪多糖组均显著高于对照组(P<0.05),培哚普利组和黄芪多糖组低于模型组(P<0.05)。③ATIR的表达:糖尿病模型组高于正常对照组(P<0.05),培哚普利组、黄芪多糖组低于模型组(P<0.05)。结论①糖尿病大鼠肺组织中肾素血管紧张素系统活性增加,可能参与了肺组织的损害。②黄芪多糖、培哚普利能降低肾素血管紧张素系统活性,改善肺结构。

Effect of astragalus polysaccharide on renin-angiotensin system activity in diabetic rat lung

Abstract:Objective To investigate the effect of astragalus polysaccharide on renin-angiotensin system activity in diabetic rat lung.Methods Heath SD rats were divided into normal group,model group,perindopril group,astragalus polysaccharide group,and were administered saline(normal and model group),perindopril 2mg/kg,astragalus polysaccharide1mg/kg,once per day,five times per week.Rats were killed after 8 weeks,lung tissue was homogenated,Angiotensin II was detected.Pathological changes in lung tissue was observed through HE staining.Expression of Angiotensin II receptor I(ATIR) was measured by immunohistochemistry.Results ① Alveolars were collapsed,destructed,alveolar septum was widened in diabetic model rats.Alveolar structure was improved in perindopril and astragalus polysaccharide group.②Angiotensin II concentration: Angiotensin II concentrations in diabetic model,perindopril and astragalus polysaccharide group were higher than that in normal group(P<0.05).Angiotensin II concentrations in perindopril and astragalus polysaccharide group were less than that in diabetic model group(P<0.05).③Expression of ATIR: The expression of ATIR in diabetic model group was higher than in normal rats(P<0.05),in perindopril and astragalus polysaccharide group was less than that in diabetic model group(P<0.05).Conclusion Renin-angiotensin system activity elevated in diabetic rat lung tissue,which may participate in the lung damage.Astragalus polysaccharide and perindopril reduced it and improved lung structure.
