全反式维甲酸对糖尿病大鼠肾小管-间质Prohibitin表达的影响

摘    要:目的探讨全反式维甲酸对糖尿病大鼠肾小管-间质Prohibitin表达的影响。方法24只链脲佐茵素(STZ)诱导的糖尿病大鼠,随机分为治疗组(T),模型组(D)各12只和12只正常组(N),T组给予全反式维甲酸20mg/(kg·d)油溶液灌胃,D组和N组分别灌等量的溶剂,4、8周每组各处死大鼠6只,测量尿清蛋白排泄率、肾重/体重、血肌酐、血糖(BS)。肾组织Masson’s染色。免疫组化方法检测肾小管-间质Prohibitin的表达。结果全反式维甲酸可缓解肾小管的损伤,减少肾间质的细胞外基质,4、8周时尿清蛋白的排泄率治疗组分别显著低于模型组(P〈O.01),血肌酐治疗组与同期模型组相比也显著降低。治疗组肾小管.间质Prohibitin的表达上调,明显高于模型组。结论全反式维甲酸可能通过上调肾小管-间质Prohibitin的表达防止肾间质的纤维化,保护肾功能。

Abstract:Objective To explore the effect of all-trans retinoic acid(atRA) on the expression of probibitin of renal tu- bule-interstitium in diabetic rats. Methods 24 diabetie rats induced by streptozotocin(STZ)were randomly divided into therapy (T) and model(D) group,in addition, 12 normal rats as control group(N). The rats in therapy group were administrated by atRA, which was dissolved by vegetable oil, at a dosagc of 20mg/( kg ~ d), D and N group were treated with vegetable oil. Excretory rate of urinary protein, the ratio of kidney and body weight, serum creatinine, BS were measured after 4 and 8 weeks. The morphological changes were observed by Masson's staining. The expression of prohibitin in renal tubule-interstitium were assessed by immunohis- tochemical. Results atRA significantly attenuated damage of renal tubule and reduced extracellular matrix of renal interstitium excretory rate of urinary protein and serum creatinine in therapy group was significany low compare to model group ( P〈0. 01 ) re- spectivcly after 4,8 weeks. The expression of prohibitin in renal tubule-interstitium in therapy group was significandy up-regulated compare to model group. Conclusion atRA up-regulated the expression of prehibitin in renal tubule-interstitium, prevented fibro- sis of renal interstitium.
