汉防己甲素对兔视网膜色素上皮细胞Bax、Bcl-2表达的影响

摘    要:目的:评价汉防己甲素(Tet)对兔视网膜色素上皮(RPE)细胞增殖的抑制作用及对凋亡蛋白Bax、Bcl-2表达的影响。方法:MTT法检测Tet对体外培养兔RPE细胞增生的影响,流式细胞仪检测Tet对兔RPE细胞增殖周期、凋亡率及Bax、Bcl-2蛋白表达的影响。结果:Tet对兔RPE细胞的抑制率均随作用时间的延长而增高,各时间点之间差异均有统计学意义(P〈0.05);抑制率均随药物质量浓度的增高而增高,除Tet10μg/ml、15μg/ml外,各质量浓度之间差异有统计学意义(P〈0.05);Tet10μg/ml作用72 h后,出现G0/G1期细胞明显增多,S期与G2/M期细胞降低,Bax蛋白表达增加,Bcl-2蛋白表达下降,Bcl-2/Bax比值下降,以上与对照组相比均有显著性差异(P〈0.05)。结论:Tet可能通过干预RPE细胞Bax与Bcl-2蛋白的表达而对其增殖产生抑制作用。

Effects of tetrandrine on expression of Bax and Bcl-2 proteins of retinal pigment epithelium cells in rabbits

Abstract:Objective:To evaluate the inhibiting effect of tetrandrine(Tet) on retinal pigment epithelium(RPE) cells and on expression of Bax and Bcl-2 proteins of RPE cells in rabbits.Methods:The inhibiting effects were detected by MTT.Flow cytometry was used to detected the effects of Tet on the proliferating cycle,the apoptosis rate,expression of Bax and Bcl-2 apoptosis proteins of RPE cells in vitro.Results:The growth inhibition ratio of RPE cells increased with the increase of Tet's action time.The ratio of each time point was statistically different(P0.05).The growth inhibition ratio increased with the increase of Tet's concentration.There was statistical difference among each group of different concentration except 10 μg/ml and 15 μg/ml group(P0.05).After 72 h in Tet 10 μg/ml group,the RPE cells statistically increased in G0/G1 period and decreased in S and G2/M periods.The expression of Bax protein increased,however Bcl-2 protein and Bcl-2/Bax ratio decreased statistically(P0.05).Conclusion:The rabbit RPE cells may be inhibited by Tet through interfering with expression of Bax protein and Bcl-2 proteins.
