| 摘 要： | 目的 探讨生长抑素类似物奥曲肽对人脐静脉内皮细胞增殖的影响及其机制。方法 以奥曲肽作用于人脐静脉内皮细胞，通过四噻氮唑盐法(MTT法)和流式细胞术进行分析。结果 在体外，奥曲肽(10^-5～10^-8mol／L)对人脐静脉内皮细胞可以产生抑制作用，并见饱和现象；10^-8mol／L时产生G0／G1期细胞阻滞，且出现细胞凋亡峰。结论 奥曲肽对人脐静脉内皮细胞具有抑制效应，影响细胞周期和诱导细胞凋亡是其作用机制之一。 |

| Abstract: | Objective To investigate the effect and mecha nism of the somatostatin analogue octreotide on the growth of human umbilical vein endothelial cell (HUVEC) cultured on gelatin-coated dishes. Methods Using MTT method and flow cytometry, the action of octreotide on the cell cycle and proliferatio n in phase-synchronized HUVEC was observed. Results Octreotide in concentrations from 10 -5 mol/L to 10 -8 mol/L diminished the proliferative activi ty of cultured HUVEC vs control's. Furthermore, the G 0/G 1-phase cell phase blockade a nd typical apoptotic peak were detectable in phase-synchronized cells subjected to treatment of octreotide. Conclusion Octreotide is an effective inhibitor of vascular cell proliferation in vitro. The induction of apoptosi s and arrest of cell cycle may be the primary mechanism. |
