舒张功能不全大鼠心肌线粒体结构和功能的改变

摘    要:目的探讨心肌线粒体在舒张功能不全心力衰竭(DHF)大鼠心脏功能损伤中的作用。方法20只雄性SD大鼠,随机分为阴性对照组和DHF组。采用腹主动脉缩窄术建立DHF模型,4周末心脏超声检测心功能,颈动脉插管记录血流动力学,分光光度计检测心肌线粒体丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSHPx)水平,电镜检测心肌线粒体超微结构。结果DHF组大鼠左心室后壁(LVPW)、室间隔(IVS)、左心室心脏指数(LVM)和E/A比值增高,主动脉收缩压(SBP)、舒张压(DBP)、左心室收缩压(LVSP)和左心室舒张末期压(LVEDP)升高,左心室松弛时间常数(Tau)延长,平均左心室内压最大下降速率(LV-dp/dtmax)下降,心肌SOD和GSHPx下降,MDA增加,电镜示心肌细胞肌丝排列不整齐、线粒体肿胀及空泡化等线粒体损伤。结论心肌线粒体损伤在DHF大鼠心脏功能损害中起着重要的作用。

Alteration of the Structure and Function in Cardiac Mitochondrian of Rats With Diastolic Heart Failure

Abstract:Objective To study the effect of cardiac mitochondrian on cardiac function of rats with diastolic heart failure. Methods 20 SD rats were randomly divided into control group and diastolic heart failure (DHF) group. DHF model was produced by abdominal aortic coarctation in the rats in the operating group. After 4 weeks, cardiac function and hemorheology was assessed by echocardiography and catheterization. Specimens were taken and ventricular mass were meas- ured. The level of malonaldehyole(MDA), superoxidedismutase (SOD), glutathioneperoxidase(GSH Px) were detected by speetrophotometer. Ultra structure changes in myocardium were observed under eleetrion microscopy. Results In DHF group, left posterior wall (LVPW), interventricular septum (IVS) ,left ventricular mass (LVM) , E/A ratio, systohc blood pressure (SBP) ,diastolic blood pressure (DBP) ,left ventricular systolic pressure (LVSP) ,left ventrieular end diastolic pressure (LVEDP) , MDA (32.6±4.2 nmol/mg vs 13.9±1.8 nmol/mg, P 〈 0.05) were increased, constant for left ventricular relaxation (Tau) was prolonged, maximum rate of left intraventricular pressure down (LV-dp/dtmax ), SOD 17.9±2.1 U/mg vs 36.9±3.5 U/mg,P 〈0.05) and GSH Px(2.5±0.2 U/mg vs 7.7±0.9 U/mg, P 〈 0.05) was decreased. The myocardium damage including abnormality of myofilament arrangement,mitochondrial swelling and vacuolization were showed. Conclusions The damage of myocardic cell and mitochondrion has an important effect on cardiac function of rats with diastolic heart failure.
