缬沙坦螺内酯及其合用对心力衰竭大鼠细胞因子胶原产生的影响

目的研究缬沙坦、螺内酯及其合用对心力衰竭大鼠细胞因子、胶原产生的影响,探讨二者合用治疗心衰的可能机制。方法采用腹主动脉缩窄法复制慢性心力衰竭大鼠模型。大鼠随机分为5组：正常组、模型组、缬沙坦组（缬沙坦20mg/（kg·d））、螺内酯组（螺内酯40mg/（kg·d））和缬沙坦＋螺内酯组（缬沙坦20mg/（kg·d）＋螺内酯40mg/（kg·d））,模型组予等容积蒸馏水灌胃。连续给药8周后,分离血清,ELISA及放免法分别检测大鼠血清中IL-6、TNF-α、BNP及PⅢNP水平。结果与模型组比较,缬沙坦组及联合给药组大鼠血清IL-6、TNF-α、BNP、PⅢNP水平均显著下降,螺内酯组大鼠血清TNF-α、BNP、PⅢNP水平均显著下降,而IL-6水平无明显改善。缬沙坦＋螺内酯联合给药组与单独给药组比较,对各项指标的抑制作用更加明显。结论缬沙坦及螺内酯对心衰大鼠的治疗作用与其抑制炎症细胞因子IL-6、TNF-α的表达,抑制BNP及PⅢNP的产生有关,联合给药对上述因子具有协同抑制作用。

The Effects of Aldactone, Valsartan and their combination on cytokines and collegen production in Rat Model of Heart Failure

Objective To investigate the effects of aldactone,valsartan and their combination on cytokines and collegen production in serum of rat model with heart failure.Methods Rat model with heart failure was induced by descending aortic constriction(DAC).60 rats were randomized into five groups as normal control group,model group,valsartan group,aldactone group,combination of valsartan and aldactone group.After drug treatment for 56 days,the tumor necrosis factor-alpha(TNF-α),interleukin-6(IL-6),BNP and PⅢNP levels in serum were measured by ELISA and radio immunoassay.Results Compared with normal group,the level of TNF-α,IL-6,BNP and PⅢNP in serum of rats in the heart failure group significantly increased(P < 0.01).Aldactone,valsartan and their combination administration significantly decreased the plasma content of TNF-α,IL-6,BNP,as well as PⅢNP.Further more,aldactone and valsartan had synergistic effect on these cytokines and collegen production.Conclusion Aldactone and valsartan administration could improve the heart function of heart failure rats,which might be due to its inhibition of TNF-α,IL-6,BNP and PⅢNP production in blood.Aldactone and valsartan had synergistic inhibition effect on cytokine and collegen production.
