妊高征胎盘组织脂代谢相关基因的异常表达

目的：了解妊娠高血压综合征患胎盘组织有关代谢基因的表达，寻找妊高征血管内皮损伤的原因．方法：选择4例重度妊高征胎盘组织为研究对象，以混合的4例正常胎盘总RNA为对照，用cy5dUTP和cy3dUTP分别标记妊高征胎盘组织cDNA和对照cDNA．将与代谢、凋亡等有关基因的cDNA点在特殊的硅化玻片上，制成4000点的cDNA表达谱芯片，用该芯片检查相关基因的表达．结果：在4次芯片杂交过程中，共有58-131条不等的基因发生差异表达，其中出现2次以上重复一致的异常表达基因共22条：表达增高的有13条，表达降低的有9条．脂代谢相关基因脂肪分化相关蛋白(ADRP)基因在3例胎盘组织中表达增高；脂蛋白脂酶(LPL)基因在4例妊高征胎盘组织中表达均降低．结论：脂代谢相关基因表达异常可能是血管内皮损伤的原因之一．

Abnormal expression of lipid metabolism- related genes in placentas of pregnancy-induced hypertension

AIM: To identify the factor(s) damaging endothelium in pregnancy induced hypertension (PIH) by testing the metabolism related gene expression difference between PIH placentas and normal ones. METHODS: 4 samples of placental tissue from 4 severe PIH patients were studied as experimental samples. mRNA levels were detected using 4,000 feature cDNA Microarray, a special silica slide on which 4000 cDNA of genes relevant to metabolism and apoptosis was spotted and compared with the pooled control consisting of total RNA from 4 cases of normal placentas after the control cDNA and experimental cDNA were labeled by cy3dUTP and cy5dUTP, respectively. RESULTS: In 4 runs of hybridization, 58 to 131 genes were found down or up regulated. Among the differentially expressed genes, 22 genes had the concordance in at least 2 cases: 13 were up regulated and 9 down regulated. Adipose differentiation related protein (ADRP) gene was found up regulated in 3 cases of PIH and lipoprotein lipase (LPL) gene down regulated in 4 cases of PIH. CONCLUSION: cDNA Microarray is a high throughput and time saving method to monitor the altered expression of genes relevant to lipid metabolism, which may be responsible for the endothelial damage.
