摘    要:目的:探讨金属硫蛋白(metallothioneins,MTs)在小鼠变应性接触性皮炎中所起的作用。方法:用二硝基氟苯(dinitrofluorobenzene,JNFB)致敏金属硫蛋白基因敲除小鼠(metallothionein-null mouse,MT-/-,不表达MT-I及MT-Ⅱ)及基因同源对照野生型鼠(MT+/+,正常表达MTs),并在小鼠右耳激发变应性接触性皮炎,分别对比两组小鼠激发后耳肿胀度。结果:野生型鼠(MT+/+)耳肿胀度明显高于基因敲除鼠(MT-/-),统计学分析差异有显著性(P<0.05,t检验)。结论:MTs在迟发型变态反应的炎症过程中起重要作用,MT-/-小鼠不能完全表达迟发型变态反应的炎症。

Abstract:OBJECTIVE: To investigate the chronic hypersensitivity (CHS) to dinitrofluorobenzene (DNFB) in metallothionein gene knocked-out (MT -/-) mice and wild mice (MT +/+) with homologous genes. METHODS: Mice were sensitized by applying DNFB on abdominal skin and CHS was induced on right ears. The CHS was observed by ear swelling. RESULTS: In comparison with metallothionein--null mice, the ear swelling of normal control wild mice was significantly greater. (P < 0.05, Students t test). CONCLUSION: Metallothioneins play an important role in the CHS. Metallothionein gene knocked-out (MT -/-) mice express less inflammation of CHS.
