| 摘 要： | 目的在体外细胞培养研究中,观察Adefovir抗HBV的作用.方法以乙型肝炎病毒(HBV)DNA转染的细胞株2.2.15细胞为靶细胞,给药后通过检测细胞培养液中HBsAg、HBeAg和HBV DNA,对Adefovir抗HBV效果进行评价.在2.2.15细胞培养基中分别加入不同浓度的药物,培养数天后检测上清液中HBsAg和HBeAg滴度,细胞DNA抽提液检测HBV DNA;用四甲基偶氮唑盐(MTT)比色分析法检测药物对细胞的毒性.结果 Adefovir加入细胞培养12d,最大无毒浓度为20μg/ml,对HBsAg抑制率为52.44%,对HBeAg抑制率为54.87%,对HBV DNA抑制率为45.68%,并有剂量依赖效应.结论 Adefovir在体外细胞培养的实验中有抗HBV病毒作用.

| Abstract: | Objective To investigate the effect of anti-HBV of Adefovir in vitro. Method The 2.2.15 cell line were treated with series concentrations of Adefovir, then the HBsAg and HBeAg in the supernatant and HBVDNA both in supernatant and cell were measured with commercial detective kits. At the same time the cell vitality also was tested with MTT method for evaluating the cyto-toxic effect of Adefovir. Result After 12d,Adefovir showed no cyto-toxic effect on 2.2.15 cell in the concentration under 20mg/ml. The inhibitive rates were 52.44% for HBsAg, 54.87 for HBeAg, and 45.68% for HBVDNA with the dose-dependent.Conclusion Adefovir shows the anti-HBV action in 2.2.15 cell line culture. |
