沙棘叶中总黄酮的提取和纯化

摘    要:目的研究不同方法对沙棘叶总黄酮的提取效果以及大孔吸附树脂对沙棘叶总黄酮的纯化性能及纯化条件。方法采用索氏提取法、超声提取法和微波提取法提取沙棘叶总黄酮,用X-5大孔吸附树脂对沙棘叶总黄酮进行纯化,并用紫外分光光度法测定其含量。结果索氏提取法提取6h的提取率为19.78%,超声提取10min的提取率为19.19%,微波提取10min的提取率为24.19%;X-5大孔吸附树脂对沙棘叶总黄酮的纯化性能较佳,其对沙棘叶总黄酮的纯化条件为:上样液浓度1.159-1.545mg/ml,上样液pH值4.06,洗脱剂为50%乙醇。结论循环超声提取10min即与索氏提取6h的提取效果相当,而微波提取技术大大缩短了提取时间,而且提取率有所增加;X-5树脂适合应用于沙棘叶总黄酮的纯化。

Abstract:Objective To study the extraction effect of total flavones from leaves of Hippophae rhamnoides L.by different methods and the adsorption function of macroporous resin for the purification of total flavones in leaves of Hippophae rhamnoides L.Methods The Soxhlet,circulative ultrasonic wave,and microwave were used for flavones extraction,X-5 resin was used to purify the total flavones,and UV spectrophotometry was used to analyze the contents of total flavones.Results The extraction rates were 19.78% with Soxhlet for 6 h,19.19%with circulative ultrasonic wave for 10 min and 24.19% with microwave for 10 min.X-5 resin had capability of selective adsorption of total flavones from the leaves of Hippophae rhamnoides L.The optimal adsorption were 1.159-1.545 mg/ml of solution concentration at pH 2,and the optimal desorption was 50% ethanol as eluting solvent.Conclusion The effect of circulative ultrasonic extraction for 10 min is almost equal to Soxhlet extraction for 6 h.And the effect of microwave extraction is even faster and more efficient.X-5 resin has favorable effect of adsorption and desorption for purifying total flavones from leaves of Hippophae rhamnoides L.
