胶原抗原肽表位诱导的耐受性抗原递呈细胞对大鼠类风湿关节炎的治疗作用

目的:观察抗原肽表位(CTE)1和CTE2诱导、TGF-β2刺激的抗原递呈细胞(APC)对大鼠类风湿关节炎的疗效.方法:雌性Wistar大鼠32只,分为关节炎组、免疫性细胞组、耐受性细胞组和正常对照组,每组8只.关节炎、免疫性细胞和耐受性细胞组大鼠使用鸡Ⅱ型胶原(CⅡ)加完全福氏佐剂诱导CIA后,分别静脉注射免疫性APC、耐受性APC和Hanks平衡盐溶液(HBSS),1周后观察大鼠关节炎指数,1月后检测外周血血清中抗CⅡ抗体吸光度(A)值,流式细胞仪检测各组大鼠外周血CD4+T细胞中Th1和Th2细胞比例.结果:各组大鼠关节炎指数、外周血抗CⅡ抗体A值和Th1型/Th2型CD4+T细胞比率比较,差异均有统计学意义(F=20.911、22.498和10.775,P＜0.001);耐受性细胞、免疫性细胞和关节炎组较正常对照组升高(P＜0.05),耐受性细胞组较免疫性细胞和关节炎组降低(P＜0.05).结论:CTE1和CTE2诱导、TGF-β2 刺激的APC对大鼠类风湿关节炎有抑制作用.

Therapeutic effect of tolerogenic antigen presenting cell on collagen-induced arthritis in rats

Aim:To observe the therapeutic effects of tolerogenic antigen presenting cell(APC) on collagen-induced arthritis rats.Methods:Female Wistar rats were divided into 4 groups:arthritis group,immunogenic APC group,tolerogenic APC group,and normal control group,and each group contained 8 rats.HBSS,immunogenic APCs,and tolerogenic APCs were injected to the rats in the first 3 groups after the establishment of arthritis model induced by chicken type Ⅱ collagen(CⅡ) with complete Freund’s adjuvant.The efficacy of this therapy was verified by clinical visual scoring and serum levels of anti-CⅡ IgG.The Th1 type to Th2 type CD4+ cells was detected by flow cytometry.Results:Clinical visual scoring of arthritis,serum levels of anti-CⅡ IgG,and the ratio of Th1 type to Th2 type cells among 4 groups had significant differences(F=20.911,22.498,10.775,P<0.001);they were higher in tolerogenic APC,immunogenic APC,and arthritis groups than those in normal control group(P<0.05),and were lower in tolerogenic APC group than those in immunogenic APC and arthritis groups(P<0.05).Conclusion:APCs treated with TGF-β2 and pulsed by CETs can suppress CIA significantly.
