抗HCV NS3抗原单克隆抗体的研制与初步鉴

目的：为研制抗丙型肝炎病毒（ＨＣＶ）ＮＳ３抗原单克隆抗体，用于检测丙肝患者血清中的ＨＣＶＮＳ３抗原。方法：用基因工程表达的ＨＣＶ非结构区ＮＳ３蛋白与鼠血清白蛋白交联后，免疫ＢＡＬＢ／ｃ小鼠，用杂交瘤技术获得能分泌单克隆抗体（ＭｃＡｂ）的细胞株。结果：获得有实用价值的两株ＭｃＡｂ。两株ＭｃＡｂ与免疫抗原有较强的抗原－抗体反应，与ＨＣＶＮＳ４、ＮＳ５及核心区Ｃ３３肽、ＣＰ９、ＣＰ１０抗原无反应性，在竞争ＥＬＩＳＡ中，对ＨＣＶ－ＩｇＧ阳性血清有较好的抑制作用。结论：用基因工程表达的ＨＣＶ非结构区ＮＳ３蛋白，能够制备出有实用价值的单克隆抗体。

Preparation of monoclonal antibodies against HCV NS3 antigen and their characteristic study

Objectives:]To prepare BALB/c mice monoclonal antibodies against HCV NS3 antigen and to detect HCV NS3 protein in hepatitis C patients sera. WTHX Methods: BALB/c mice were immunized with HCV NS3 antigen which was expressed by gene cloning and coupled with murine serum albumin,and the monoclonal antibodies were obtained by hybridoma technique. Results: Two monoclonal antibodies( 2B8, 6D6) were obtained.The reactions of the 2 McAbs were strong with the immunized antigen NS3,but no reactivities were detectable with HCV NS4,NS5 and its core antigen C33 peptide,CP9 and CP10.The 2 McAbs could inhibit antiHCVIgG positive serum in the competitive ELISA. Conclusion: From gene cloning HCV NS3,two monoclona antibodies against HCV NS3 protein were prepared with practical value.
