芪莲化积合剂对肝癌H22荷瘤小鼠抑瘤作用及miRNA122表达的影响

摘    要:目的观察芪莲化积合剂对肝癌H22荷瘤小鼠的抗肿瘤作用。方法建立肝癌H22荷瘤小鼠实体瘤模型,将24只小鼠分为3组,对照组(生理盐水组)、芪莲化积合剂组和5-氟脲嘧啶(5-Fu)组各8只。通过测定荷瘤小鼠的瘤重、抑瘤率观察芪莲化积合剂剂对肿瘤生长的影响,采用RT-PCR技术检测miRNA122的表达情况。结果芪莲化积合剂能使肝癌H22荷瘤小鼠的瘤重减少,显著低于对照组(P<0.05);胸腺指数、脾指数的比较,芪莲化积合剂较对照组明显升高(P<0.05);在miRNA122的表达上,芪莲化积合剂较对照组明显升高(P<0.05)。结论芪莲化积合剂具有明显的抗肿瘤作用,其作用机制可能与miRNA122的表达有关。

Abstract:Objective To observe the anti-tumor effect of Qilian Huaji mixture on mice with hepatocellular carcinoma(H22).Methods A solid tumor model of H22 tumor bearing mice was established,and 24 mice were divided into three groups:8 mice in normal saline group(control group),8 mice in qilianhua mixture group and 8 mice in 5-fluorouracil(5-Fu)group.The effect of qilianhua mixture on tumor growth was observed by measuring tumor weight and tumor inhibition rate in tumor-bearing mice,and the expression of miRNA122 was detected by RT-PCR.Results Qilian Huaji mixture could reduce the tumor weight of H22 tumor bearing mice,which was significantly lower than that of the control group(P<0.05).Comparison of thymus index and spleen index showed that qilianhua mixture was significantly higher than the control group(P<0.05).The expression of miRNA122 was significantly higher than that of the control group(P<0.05).Conclusion Qilian Huaji mixture has obvious anti-tumor effect,and its mechanism may be related to the expression of miRNA122.
