活血化瘀中药对糖尿病肾病大鼠肾脏保护及血清降钙素基因相关肽的影响

摘    要:目的研究活血化瘀中药对糖尿病肾病大鼠肾脏保护及血清降钙素基因相关肽(CGRP)的影响。方法 SD大鼠分为正常组(n=16)、模型组(n=16)、中药组(n=16),模型组和中药组利用单侧肾切除加腹腔注射链脲佐菌素(STZ)造成糖尿病肾病大鼠模型。正常组和模型组灌服等容积蒸馏水。中药组在单肾切除STZ诱导糖尿病肾病模型成模后灌胃活血化瘀中药。第8、12周末分2批检测24 h尿蛋白定量(24h UTP)、血糖(Glu)、血尿素氮(BUN)、肌酐(Cr)及体质量的变化,放射免疫法测定CGRP。结果第8、12周末模型组24 h UTP、Glu、体质量与正常组比较差异均有统计学意义(P0.05);第8、12周末中药组24 h UTP与模型组比较差异有统计学意义(P0.05);第8、12周末中药组Glu与模型组相比有下降趋势,但无统计学意义(P0.05);第8、12周末中药组体质量与模型组比较差异有统计学意义(P0.05);第12周与第8周相比,中药组体质量有上升趋势,差异有统计学意义(P0.05)。第8、12周末模型组BUN与正常组同期比较差异有统计学意义(P0.05);第12周末中药组BUN与模型组相比有下降趋势,第12周与第8周相比中药组下降,差异有统计学意义(P0.05);随着造模时间的延长,模型组血Cr含量比正常组稍有增加,但无统计学意义(P0.05),第12周末中药组与模型组相比有下降趋势,无统计学意义(P0.05);第8、12周末模型组和中药组的CGRP升高,与正常组同期比较差异均有统计学意义(P0.05),且中药组CGRP与模型组同期比较差异有统计学意义(P0.05),第12周与第8周中药组CGRP比较升高,差异有统计学意义(P0.05)。结论活血化瘀中药对单肾切除STZ诱导糖尿病肾病大鼠有降低蛋白尿,调节血糖代谢,保护肾功能的作用。活血化瘀中药对单肾切除STZ诱导糖尿病肾病大鼠能明显扩张肾脏血管,增加肾脏血流量,改善肾脏血液微循环,有效提高肾脏滤过率,减轻肾脏病理损害,从而达到保护肾功能的作用。

Effects of blood activating and stasis - removing Chinese medicine on kidney protection and serum calcitonin generelated peptide in rats with diabetic nephropathy
Abstract:Objective To observe the effects of blood activating and stasis - removing Chinese medicine on kidney protection and serum calcitonin generelated peptide(CGRP)in rats with diabetic nephropathy(DN). Meth-ods The SD rats were divided into normal group(n = 16),DN model group(n = 16),Chinese medicine group(n= 16). The model and Chinese medicine group were established by uninephrectomy and intraperitoneal injection of streptozotocin. Rats in normal and model group received equivalent distilled water. The Chinese medicine group re-ceived blood activating and stasis - removing Chinese medicine by gavage after the DN models established successful-ly. The 24 hour urine proteinuria(24 hUTP),blood glucose(Glu),blood urine nitrogen(BUN),creatinine(Cr) and body weight at the 8 weeks and 12 weeks of treatment were measured. The CGRP was measured by radioimmuno-assay. Results There were statistical differences on 24 hUTP,Glu and body weight after 8 weeks and 12 weeks of treatment between model and normal groups(P < 0. 05). There were statistical differences on 24 hUTP after 8 weeks and 12 weeks of treatment between Chinese medicine and model groups(P < 0. 05). The Glu after 8 weeks and 12 weeks of treatment in Chinese medicine group with a downward tendency as compared with that in model group,but with no statistical difference(P > 0. 05). There were statistical differences on body weight after 8 weeks and 12 weeks of treatment between Chinese medicine and model groups(P < 0. 05). The body weight at the 8 weeks and 12 weeks showed an upward tendency in Chinese medicine group,with statistical difference(P < 0. 05). There were statistical differences on BUN after 8 weeks and 12 weeks between model and normal group(P < 0. 05). The BUN at the 12 weeks in Chinese medicine group showed a downward tendency as compared with that in model group,and the BUN in Chinese medicine group showed a downward tendency between the 12 weeks and the 8 weeks,with statistical differences(P < 0. 05). As the modeling time went on,the Cr in model group was higher than normal group,but with no statistical difference(P > 0. 05). The Cr at the 12 weeks in Chinese medicine group showed a downward tendency as compared with that in model group,with no statistical difference(P > 0. 05). The CGRP at 8 and 12 weeks were increased in model and Chinese medicine group as compared with those in normal group at the same peri-od,with statistical differences(P < 0. 05). There were statistical differences on CGRP between Chinese medicine and model group at the same period(P < 0. 05). The CGRP at 12 weeks in Chinese medicine group was increased as compared with the 8 weeks(P < 0. 05). Conclusion Blood activating and stasis - removing Chinese medicine can decrease proteinuria,regulate glycometabolism and protect kidney function in mono - nephrectomy STZ - induced DN rats. Blood activating and stasis - removing Chinese medicine can obviously dilate vessel of kidney,increase kidney blood flow,effectively enhance filtration rate,and reduce kidney damage,thus achieve the protection of kidney func-tions.
