木香对脾虚大鼠胃肠道运动及血清乙酰胆碱酯酶、一氧化氮水平的影响

摘    要:目的观察木香对脾虚大鼠胃肠道运动及血清乙酰胆碱酯酶(AChE)、一氧化氮(NO)水平的影响。方法将92只SD大鼠随机分为正常对照组、莫沙必利对照组、木香生品高剂量组、木香生品中剂量组、木香生品低剂量组、木香麸煨品高剂量组、木香麸煨品中剂量组及木香麸煨品低剂量组,每组10只,模型对照组12只。除正常对照组外,其余各组大鼠均通过腹腔注射利血平建立脾虚模型。造模成功后,木香生品高、中、低剂量组和木香麸煨品高、中、低剂量组分别按照7.00、3.50、1.75 g/(kg·d)的浓度予相应药液灌胃,莫沙必利对照组予枸橼酸莫沙必利分散片0.35 mg/(kg·d)药液灌胃,正常对照组及模型对照组(模型对照组取其中10只)分别予0.9%氯化钠注射液7.00 mg/(kg·d)灌胃。连续灌胃14 d后,比较各组大鼠血清AChE及NO水平变化情况。将模型对照组剩余2只大鼠采用脱颈法处死,解剖后将十二指肠取出,剪成数小段,放入离体肠管活动生理记录装置中,记录其收缩曲线,计算平均振幅,然后依次加入木香生品和木香麸煨品高、中、低浓度药液1.0、0.5、0.25 g/mL,比较各组大鼠离体十二指肠相对收缩振幅情况。结果与正常对照组比较,模型对照组灌胃后血清NO含量明显升高(P<0.05),AChE含量明显下降(P<0.05)。与模型对照组比较,莫沙必利对照组灌胃后血清NO含量明显下降(P<0.05),AChE含量明显升高(P<0.05),而木香生品和麸煨品各剂量组中仅木香生品高剂量组和木香生品中剂量组灌胃后AChE含量明显升高(P<0.05),其余比较差异均无统计学意义(P>0.05)。与模型对照组比较,木香生品高剂量组大鼠离体十二指肠相对收缩振幅明显增强(P<0.05),其余比较差异均无统计学意义(P>0.05)。结论木香对胃肠运动具有兴奋作用,可明显促进胃肠道运动,其中以木香生品高剂量组效果最明显,这也是木香理气作用的体现,其作用机制可能与升高ACh水平有关,但对NO作用不明显。

Abstract:Objective To observe the effect of Muxiang on gastrointestinal motility and serum acetylcholinesterase(AChE)and nitric oxide(NO)levels in rats with spleen deficiency.Methods 92 SD rats were randomly divided into normal control group,mosapride control group,muxiang crude high dose group,muxiang crude middle dose group,muxiang crude low dose group,muxiang simmer high dose group,muxiang simmer middle dose group and muxiang simmer low dose group,with 10 rats in each group and 12 rats in model control group.In addition to the normal control group,other groups of rats were injected with reserpine intraperitoneally to establish the spleen deficiency model.After the model was established successfully,the high,middle and low dose groups of muxiang crude and the high,middle and low dose groups of muxiang simmer were given the corresponding solutions respectively according to the concentrations of 7.00,3.50 and 1.75 g/(kg·d)by gavage.The mosapride control group was given 0.35 mg/(kg·d)of mosapride citrate dispersible tablet solution by gavage.The normal control group and model control group(10 of them in model control group)were given 7.00 mg/(kg·d)0.9%sodium chloride injection by gavage.The changes of serum AChE and NO levels in each group were compared after 14 days of continuous gavage.The remaining two rats in the model control group were killed by the method of neck removal.After dissection,the duodenum was taken out and cut into several small segments,which were put into the physiological recording device of isolated intestinal tract activity,the contraction curve was recorded,the average amplitude was calculated,and then the high,middle and low concentration solution(1.0,0.5,0.25 g/mL)of crude Muxiang and simmered Muxiang were added successively,and the relative contractile amplitude of isolated duodenum was compared.Results Compared with the normal control group,the NO content of the model control group was increased significantly(P<0.05),and the AChE content was decreased significantly(P<0.05).Compared with the model control group,the NO content and AChE content of mosapride control group were decreased significantly(P<0.05),and the AChE content was increased significantly(P<0.05).However,in each dose group of crude Muxiang and simmered Muxiang,only the high dose group and the middle dose group increased the AChE content significantly(P<0.05),and there was no significant difference in other comparisons(P>0.05).Compared with the model control group,the relative contraction amplitude of isolated duodenum only increased significantly in the high dose group of Muxiang crude(P<0.05),and there was no significant difference in other comparisons(P>0.05).Conclusion Muxiang can stimulate gastrointestinal motility and promote gastrointestinal motility,especially in the high dose group.It is also the embodiment of the effect of Muxiang on regulating Qi.Its mechanism may be related to the increase of AChE level,but it has no obvious effect on No level.
