摘    要:目的:考察大孔树脂富集牡丹皮中芍药苷的工艺参数.方法:以芍药苷为指标,HPLC测定指标成分含量,采用静态、动态吸附-解吸试验考察大孔树脂型号并优选其富集纯化工艺条件.结果:采用AB-8型大孔树脂,上样液质量浓度0.125 g·mL-1,上样量2.25 BV,上样液pH 4,吸附流速2 BV·h-1,8 BV水洗除杂,14 BV 20%乙醇洗脱芍药苷,洗脱流速4BV·h-1.此条件下,芍药苷洗脱率达90.40%.结论:该优选工艺可较好的富集牡丹皮中有效成分芍药苷,可推广于工业化大生产.

Abstract:Objective: To investigate enrichment technology parameters of paeoniflorin from Paeonia suffruticosa by macroporous resin. Method: With paeoniflorin as index,which was determined by HPLC,model of macroporous resin was optimized by static and dynamic adsorption-desorption test,and to optimize its enrichment and purification technology conditions. Result: Optimal model of macroporous resin was AB-8 type,the mass concentration of sample liquid 0.125 g·mL-1,sample volume 2.25 BV,pH of sample liquid 4,adsorption flow rate 2 BV·h-1,washed impurity with 8 BV water,paeoniflorin was eluated with 14 BV 20% ethanol,desorption flow rate 4 BV·h-1.Under these conditions,elution rate of paeoniflorin was up to 90.40%. Conclusion: This optimized process could be well enrichment of paeoniflorin from P. suffruticosa,it could be promoted in industrial production.
