补脾益气方药对脾虚哮喘大鼠肺组织IκB/NF-κB信号途径的影响

目的探讨脾虚型哮喘大鼠气道炎症,血液和支气管肺泡灌洗液（BALF）中嗜酸性粒细胞百分比,肺组织核因子-κB抑制蛋白α（IκBα）表达和核因子-κB（NF-κB）活性的变化及补脾益气方药对其的影响。方法将30只雄性Wistar大鼠随机分为3组：正常对照组（A组）、脾虚型哮喘组（B组）、补脾益气方药组（C组）,采用HE染色检测肺组织病理变化,采用免疫组织化学方法检测肺组织IκBα表达水平和NF-κB p65的活性变化。结果B组与A组比较,肺组织IκBα表达显著降低,而NF-κB p65的活性显著增高（P〈0.01）;C组与B组比较,肺组织IκBα表达显著增高,而NF-κB p65的活性显著降低（P〈0.01）。结论补脾益气方药能有效增加脾虚型哮喘大鼠肺组织IκBα的表达,抑制其肺组织NF-κBp65的活性,减轻哮喘时的气道炎症变化,从而对脾虚型哮喘起到治疗作用。

Influence of Herbs of Reinforcing Spleen to Replenishing Qi on I кB/NF-кB Signaling in Rats with Bronchial Asthma of Spleen Deficency

Objective To investigate airway inflammation,number of eosinophils in blood and BALF,the expression of inhibitor protein α（I κB α） and nuclear factor-κB（NF-κB） activity in lung of rat with spleen deficiency asthma,and the effect of herbs of reinforcing spleen to replenishing Qi.Method Thirty rats were randomly divided into three groups：control group（group A）,spleen deficiency asthma group（group B）,decoction group（group C）.The pathological changes of lung was detected by HE staining,and the expression of I-κB a was significantly increased, and activity of NF-κB p65 was significantly lowered in group C （P 〈0.01）. Conclusion Herbs of reinforcing spleen to replenishing qi can effectively played treatment of asthma in rats of spleen deficiency by increasing lung tissue I-κB a expression, inhibiting NF-κB p65 activity and reducing airway inflammation.
