摘    要: 目的建立木霉素的气相色谱含量测定方法。方法色谱柱HP-5弹性石英毛细管柱(以5%苯基甲基硅氧烷为固定液,30 m×320μm,0.25μm);检测器:氢火焰离子检测器(FID);检测器温度270℃;进样口温度250℃;柱温度:程序升温,初始温度100℃,保持5 min,以10℃·min-1升温至260℃保持5 min;载气:氮气;流速1.5 mL·min-1。结果该色谱条件下,木霉素能得到很好的分离,定量法的线性范围为0.01~0.5 g·L-1,平均回收率为99.76%,相对标准偏差为1.69%。结论该气相色谱法测定木霉素准确、可靠;测定方法简单、快速、重复性好。

Abstract:OBJECTIVE To establish an identification and quantification method in fermentation by GC.METHODS The trichodermin was determined by GC with HP-5 capillary column(5% phenyl methyl siloxane,30 m×320 μm,0.25 μm) with FID detector and nitrogen as the carries gas.The injector temperature was 250 ℃,the detector temperature was 270 ℃,the column temperature was increased by program,the initial temperature was 100 ℃,maintained for 5 min,raised to 260 ℃ with a rate of 10 ℃·min-1,maintained for 5 min.The flow rate of N2 was 1.5 mL·min-1.RESULTS Trichodermin was completely separated by GC,the linearity was in the range of(0.01~0.5) g·L-1,average recovery was 99.76%,RSD was 1.69%.CONCLUSION The method of GC was accurate,simple,rapid and producible.
