摘    要: 目的研究一种测定穿山龙中薯蓣皂苷元时快速提取薯蓣皂苷元的新方法——水解原位萃取法。方法对影响提取率的重要因素进行了考察,以期建立最佳提取条件和适宜的纯化方法。结果样品用含约1.5 mol·L-1硫酸的70%异丙醇水溶液在沸水浴中加热回流4h,获得了比传统方法更高的提取率。结论水解原位萃取法简单、省时、准确。

Abstract:OBJECTIVE To study a new extraction method for analyzing diosgenin in Dioscorea nipponica Makino.---extraction in combination hydrolysis in situ.METHODS The effective factors on the yield of diosgenin were evaluated in order to establish the optimum conditions for extraction in combination hydrolysis in situ and the suitable method for isolating diosgenin from the crude extracts. RESULTS Hydrolyses was conducted in 1.5 mol·L-1 sulfuric acid with 70% isopropanol at 100℃ for 4 h. The higher recovery of diosgenin was obtained by the extraction in combination hydrdysis in situ than that by traditional methods.CONCLUSION The method is simple, accurate and time saving.
