白附子提取物抑制人胃癌 SGC-7901 细胞增殖及诱导凋亡的机制研究

摘    要: 目的 探讨白附子提取物对人胃癌 SGC-7901 细胞增殖抑制和凋亡诱导作用及其可能机制。 方法 分别采用噻唑蓝( MTT )法检测细胞增殖活性、流式细胞仪检测细胞周期及凋亡、光镜观察细胞结构变化、 Hoechst33342/PI 荧光染色检测细胞凋亡、 Western blotting 法检测 caspase-3 、 Bax 和 Bcl-2 蛋白表达。 结果 0.072~7.2 mg · L-1 白附子提取物处理 24 h 后,细胞增殖明显受到抑制,且抑制率随时间延长而增加;光镜下可观察到细胞凋亡形态的改变; Western blotting 结果显示,半胱氨酸蛋白酶( caspase-3 )酶原蛋白的激活, Bcl-2 蛋白表达的降低和 Bax 蛋白表达上调。 结论 白附子提取物通过影响 SGC-7901 细胞的增殖、周期和凋亡相关蛋白的表达发挥凋亡诱导作用。

Abstract:OBJECTIVE To explore the anti-proliferation and apoptosis-induced effects of Rhizoma Typhonii extracts on SGC-7901 cells, and possible mechanisms. METHODS SGC-7901 cell proliferative activity was measured by MTT assay. Cell cycle and apoptosis were analyzed by flow cytometry(FCM). Cell apoptosis was observed by Hoechst33342/PI fluorescent staining. The protein levels of caspase-3, Bax and Bcl-2 were detected by Western blotting. RESULTS After exposure to Rhizoma Typhonii extracts at concentrations from 0.072 to 7.2 mg·L-1 for 24 h,cellular proliferation was inhibited with the increase of the concentration and the time. The significant apoptotic morphology changes were observed. Western blotting showed the activity of the caspase-3, increase of expression of bax protein, and decrease of expression of bcl-2 protien. CONCLUSION Rhizoma Typhonii extracts induce apoptosis by up-regulation of bax protein expression and down-regulation of bcl-2 protein in SGC-7901 cells.
