子宫内膜异位症模型小鼠胞饮突时间位点观察

摘    要:目的:建立昆明小鼠子宫内膜异位症模型,选取胞饮突电镜观察的最佳时间位点。方法:以同系小鼠作为供体,取子宫内膜采用腹腔及皮下注射法对20只小鼠建立子宫内膜异位症模型。以雄性不育小鼠以2∶1合笼,次日8∶00见阴栓者为0 h。于第80 h、84 h、85 h、85.5 h、86.5 h、87 h、88 h、91 h、95 h及未假孕的小鼠,取子宫内膜行电镜观察。结果:造模成功率为90%,镜下可见第86.5 h的胞饮突生长饱满。结论:通过建立子宫内膜异位症昆明小鼠模型,成功率较高,且简单易行。胞饮突是子宫内膜容受性的重要形态学标记,它的出现与内膜种植窗口期相一致。电镜观察:在见阴栓86.5 h后,胞饮突发育完全;在见栓后80 h、84 h、85 h和85.5 h胞饮突处于发育中;在见栓后87 h、88 h、91 h和95h胞饮突逐渐退化。

Abstract:Objective: To establish the endometriosis( EMs) model in mouse and to select the best time site on Pinopodes scanning electron microscopy. Methods: With homologous mouse as a donor,20 mice with endometriosis were established by intraperitoneal and subcutaneous injection of endometrium. After two weeks,with male infertility ligation mice cage by 2: 1,0 h was defined as the day of vaginal plug at 8: 00. After examining vaginal plug,the expression of endometrial pinopodes was observed with scanning electron microscopy at 80 h,84 h,85 h,85. 5 h,86. 5 h,87 h,88 h,91 h,95 h and no false pregnancy. Results: The success rate of endometriosis model establishment was 90%. The developed pinopodes were expressed in endometrial surface at 86. 5 h.Conclusion: The success rate of endometriosis model establishment in mice with intraperitoneal and subcutaneous injection of endometrium is high and simple. The developed pinopodes are expressed in endometrial surface at 86. 5 h. The developing pinopodes are expressed at 80 h,84 h,85 h,85. 5 h. The degenerating pinopodes were expressed at 87 h,88 h,91 h,95 h. Pinopodes are the important morphologic marker of endometrial receptivity and their appearance on the surface of endometrium takes place during the implantation window.
