穴位埋线对慢性萎缩性胃炎大鼠胃黏膜及血清炎症因子表达的影响

摘    要:目的:观察穴位埋线对慢性萎缩性胃炎(CAG)模型大鼠胃黏膜及血清炎症因子表达的影响,探讨其改善CAG的机制。方法:将60只成年SD雄性健康大鼠随机分为空白组12只与造模组48只,采用自由饮用100μg/mL的N-甲基-N’-硝基-N-亚硝基胍(MNNG)溶液,并配合饥饱失常法造模,造模成功后随机分为埋线组10只、模型组9只、自然恢复组10只,模型组大鼠予以立即剖杀,采取相关标本以备检,自然恢复组、空白组不予干预措施,埋线组大鼠予以"足三里"(双)、"脾俞"(双)、"中脘穴"埋线治疗,每10 d治疗1次,共治疗6次,各组均正常饲养。采用肉眼和HE染色观察各组大鼠胃黏膜、体质量,采用ELISA法检测各组大鼠血清白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)表达。结果:①肉眼、光镜下各组大鼠胃黏膜有显著性差异,与自然恢复组相比,埋线组改善明显;②与自然恢复组相比,埋线组大鼠体质量增加明显(P0.05);③与空白组相比,模型组、自然恢复组IL-1β、IL-6、TNF-α表达均升高(P0.01);④与自然恢复组相比,埋线组IL-1β、IL-6、TNF-α表达均降低(P0.01);⑤与模型组相比,自然恢复组IL-1β、IL-6、TNF-α表达稍升高,差异无统计学意义(P 0.05)。结论:穴位埋线疗法可能通过降低炎症因子IL-1β、IL-6、TNF-α的表达从而起到抗胃黏膜损伤的作用。

Effects of Embedding Catgut in Acupoint on Gastric Mucosa and Expression of Inflammatory Factors in Chronic Atrophic Gastritis Model Rats

Abstract:Objective:To observe the effects of embedding catgut in acupoint on gastric mucosa and expression of serum inflammatory factors in chronic atrophic gastritis(CAG) model rats,and to explore the mechanism of it in improving CAG. Methods:A total of 60 male SD rats were randomly divided into the normal control group with 12 rats and the model group with48 rats. In the model group,rats drank prepared Nmethyl-N'-nitro-N-nitrosoguanidine(MNNG) solution freely every day(100 μg/mL),and the food intake was irregular. After the CAG model was established,rats were randomly divided into the embedding group(10 rats),model group(9 rats) and natural recovery group(10 rats). The rats in embedding group were treated by embedding catgut in the points of zu san li(ST 36),zhong wan(RN 12) and pi shu(DL 20) every 10 days,6 times in total,the rats in the model group were executed and the tissue samples were prepared for the further tests,and in the natural recovery group and normal group,there was no treatment. All rats were fed under normal conditions. The gastric mucosa and body weight of the rats were observed with naked eyes and also observed after hematoxylin and eosin(HE) staining under microscope. Enzyme-linked immunosorbent assay(ELISA) was used to test the expression of the related inflammatory factors like interleukin 1β(IL-1β),interleukin 6(IL-6) and tumor necrosis factor-α(TNF-α). Results:① On the observation of gastric mucosa with naked eyes or under the microscope,there was significant difference among each groups,and in the embedding group,the condition of mucosa was much better than that in the natural recovery group. ②Compared with the natural recovery group,the body weight of the embedding group was significantly increased(P<0.05). ③Compared with the normal group,the expression of IL-1β,IL-6 and TNF-α of the model group and natural recovery group was increased(P<0.01). ④Compared with the natural recovery group,the expression of IL-1β,IL-6 and TNF-α of the embedding group was decreased(P<0.01). ⑤Compared with the model group,the expression of IL-1β,IL-6 and TNF-α of the natural recovery group was slightly increased,with no statistical difference(P> 0.05). Conclusions:Embeding catgut in acupoint has effect against mucosa injury probably through reducing the expression of inflammatory factors like IL-1β,IL-6 and TNF-α.
