| 摘 要： | 目的观察防风(RS)对大小鼠胃肠运动的抑制作用并探讨其机制。方法采用苯酚红测定法观察RS对小鼠胃肠运动的影响;制备大鼠结肠平滑肌肌条并放置在浴槽中,滴加RS及其他试剂并记录肌条的收缩曲线加以分析;分离并培养大鼠结肠平滑肌细胞,探针孵育后,不同缓冲液条件下测定NS及RS处理前后细胞的荧光值并加以分析。结果 RS各组对小鼠小肠推进和胃排空均有抑制作用;RS各组均抑制离体大鼠结肠平滑肌肌条收缩,抑制Ach引起的肌条收缩。RS剂量与峰高和同线下面积有量效关系。不同缓冲液对RS处理后的荧光值和抑制率有显著性差异。结论防风可抑制小鼠小肠推进及胃排空,抑制离体大鼠结肠平滑肌收缩,机制与肾上腺素能α受体及M胆碱受体有关,与阿片受体和β受体无关;防风可降低细胞内游离钙离子浓度,其机制与抑制细胞外钙离子内流有关,与抑制细胞内钙离子释放无关。
