摘    要:目的了解慢性支气管炎大鼠模型肺与结肠病理变化关系,揭示"肺病及肠"病理传变的可能分子机制。方法采用改良烟熏法制备慢性支气管炎大鼠模型,观察正常对照组(对照组)与慢性支气管炎模型组(模型组)大鼠粪水含量;采用免疫组化法观察肺与近端结肠水通道蛋白4(AQP4)表达变化;采用ELASA法检测大鼠血浆、肺泡灌洗液及肺与近端结肠组织匀浆中血管活性肠肽(VIP)及P物质(SP)的含量。结果 与对照组相比,模型组大鼠粪水含量增加,近端结肠AQP4表达显著下降;对照组与模型组大鼠血浆、肺泡灌洗液及肺与结肠组织匀浆中VIP与SP的含量表达无差异。结论 AQP4可能是"肺液"与"肠津"联系的分子生物学基础。

Abstract:Objective:To investigate the relationship between the lung and the large intestine of the chronic bronchitis rats and to reveal the possible mechanism of the pathological transfer in the disease of lung affecting large intestine.Methods:The chronic bronchitis models were establised by the modified smoke-induced method.The feces water liquid content of the control group and the model group were observed respectively.The AQP4 expression changes of the lung and the proximal colon were detected by the immunohistochemistry.Levels including VIP and SP were determined by the ELASA in the bronchoalveolar lavage fluid and lung and proximal colon tissue homogenates.Results:Compared with the control group,the faces liquid water contents of the model group were increased,but the proximal colon of AQP4 expressions were decreased.There were no statistical differences between the control group and the model group in the content of VIP and SP of plasma,bronchoalveolar lavage fluid and lung and colon tissue homogenates.Conclusion:AQP4 may be the molecular basis of lung being connected with large intestine.
