甘草中抑制脂多糖诱导小鼠RAW264.7产生NO的活性成分研究

摘    要:目的研究甘草Glycyrrhizae Radix et Rhizoma中的抗炎活性成分。方法采用大孔树脂、ODS、半制备液相等色谱手段进行分离纯化,并通过LC-MS、1H-NMR、13C-NMR等波谱技术进行结构鉴定。用细菌脂多糖(LPS)诱导小鼠巨噬细胞RAW264.7炎症模型对分离到的化合物进行抗炎活性筛选。结果从甘草中分离得到10个化合物,分别鉴定为甘草苷(1)、芹糖甘草苷(2)、异甘草苷(3)、芹糖异甘草苷(4)、sophoraisoflavone A(5)、粗毛甘草素F(6)、光甘草酮(7)、光甘草定(8)、甘草黄酮醇(9)和粗毛甘草素D(10)。结论化合物1、3、5、6、8和9对LPS诱导的RAW264.7细胞NO分泌有一定的抑制作用。其中化合物5、6和9抑制LPS诱导的RAW 264.7细胞NO分泌为首次报道。

Abstract:Objective To study the anti-inflammatory constituents in Glycyrrhizae Radix et Rhizoma.Methods The compounds were isolated and purified by means of macroporous resin,ODS column chromatography,and semi-preparative HPLC.And their structures were identified by LC-MS,1H-NMR,and 13C-NMR.The anti-inflammatory activities of the compounds were evaluated on LPS-induced NO production in RAW 264.7 macrophages.Results Their structures were identified as liquiritin (1),liquiritin apioside (2),isoliquiritin (3),isoliquiritin apioside (4),sophoraisoflavone A (5),glyasperin F (6),glabrone (7),glabridin (8),licoflavonol (9),and glyasperin D (10).Conclusion Compounds 1,3,5,6,8,and 9 could inhibit NO production in RAW 264.7 macrophages to some extent.And the anti-inflammatory effects of compounds 5,6,and 9 are first reported in this work.
