摘    要:目的:研究黄芩汤对人结肠癌SW620细胞凋亡的作用及其机制。方法:CCK-8和MTT法检测细胞增殖;流式细胞术检测细胞凋亡;Western blot方法检测Bcl-2、Bax蛋白表达;酶标免疫测定Caspase-3和Caspase-8的相对活性。结果:与对照组相比,不同浓度黄芩汤处理的细胞增殖抑制率显著增加(P<0.05或P<0.01),细胞凋亡率升高(P<0.05或P<0.01),抑制细胞凋亡蛋白Bcl-2表达降低而促细胞凋亡蛋白Bax表达明显增高(P<0.05或P<0.01),Caspase-3和Caspase-8活性增强(P<0.05或P<0.01),并呈量效依赖关系。结论:黄芩汤能够有效的促进结肠癌SW620细胞凋亡,其机制可能与抑制Bcl-2表达而促进Bax表达并增强Caspase的活性有关。

Abstract:This study was aimed to investigate the effect of Huang-Qin-Tang (HQT) on apoptosis in human colon carcinoma SW620 cells and its mechanism. CCK-8 and MTT assay were used in the detection of cell proliferation. Flow cytometry was used to detect cell apoptosis. Western blot was used to detect expressions of Bax and Bcl-2 protein. Enzyme-linked immunosorbent assay (ELISA) was used in the determination of caspase-3 and caspase-8 activity. The results showed that compared with the control group, the inhibitory rate on cell proliferation was obviously increased after processed by different concentrations of HQT (P<0.05, or P<0.01); the cell apoptosis rate was increased (P<0.05, or P<0.01); the expression of Bcl-2, which inhibited cell apoptosis, was suppressed, but the expression of Bax, which promoted cell apoptosis, was elevated (P<0.05, or P<0.01); the activities of caspase-3 and caspase-8 were increased (P<0.05, or P<0.01). It showed the dose-dependent relation. It was concluded that HQT can effectively promote the cell apoptosis in colon carcinoma SW620 cells. Its mechanism may be related to the inhibition of Bcl-2 expression in order to promote Bax expression and strengthen caspase activity.
