不同造模方法对PCOS模型大鼠卵巢形态结构及IGF-1、IGFBP-1表达的影响

摘    要:目的:采用不同方法诱导SD清洁级雌性大鼠多囊卵巢综合征(PCOS)动物模型,造模完成后检测大鼠卵巢形态及血清中IGF-1、IGFBP-1并探讨其意义。方法:分别采用来曲唑、硫酸普拉睾酮钠、硫酸普拉睾酮钠联合HCG诱导大鼠PCOS模型,HE染色后光镜下观察卵巢局部形态,应用免疫组化法检测IGF-1、IGFBP-1在大鼠卵巢组织中的表达。结果:IGF-1在模型组大鼠卵巢组织中的表达增高,差异有显著统计学意义(P0.01);IGFBP表达在模型组大鼠卵巢组织中均降低,差异有统计学意义(P0.01,P0.05)。3种造模方法均能诱导PC0S动物模型,无论在影响IGF-1、IGFBP-1因子还是卵巢局部形态学改变方面与临床表现很接近,符合动物PCOS造模要求。

Abstract:Objective:Different methods were used to induce PCOS model,and then ovary shape and the expressions of IGF-1and IGFBP-1 were examined.Methods: Letrozol,sulfuric acid pula testosterone sodium and sulfuric acid pula testosterone sodium combined with HCG were used to induce the PCOS model.After HE staining,the ovary partial shape was observed under the light microscope,the expressions of IGF-1 and IGFBP-1 were examined by immunohistochemistry method.Results:IGF-1 expression in the model group big was increased with statistics significance(P<0.01).The IGFBP expression reduced in the model group and the difference had statistics significance(P<0.01,P<0.05).Three kinds of methods all can induce the PCOS model,both of affecting IGF-1 and IGFBP-1 or the ovary partial morphology changes are very close to the clinical manifestations,meeting the request of PCOS model.
