中药制黄精对慢性肾衰大鼠血液动力学的影响

摘    要:目的:观察制黄精对慢性肾衰大鼠血液动力学的影响,从而探讨其对糖尿病肾病大鼠肾脏的保护作用机制。方法:取清洁级SD雄性大鼠21只,随机分为假手术组及模型组,通过5/6肾切除法制造慢性肾功能衰竭(CRF)动物模型,术后2周行大鼠断尾采血,测定血清肌酐(Scr)、尿素氮(BUN)值,根据Scr随机分为病理组、制黄精组,并设假手术组。饲养或治疗8周后处死大鼠,测定血红蛋白(Hb)、红细胞(RBC)、白细胞(WBC)、血肌酐(Cr)、血尿素氮(BUN)、24h尿蛋白定量(Pro),血管内皮素(ET)、降钙素基因相关肽(CGRP)、血管紧张素Ⅰ(ATⅠ)、血管紧张素Ⅱ(ATⅡ)、TXB2、6-Keto-PGF1а。结果:制黄精组能有效改善慢性肾衰竭大鼠肾功能、血常规。与病理组相比较,制黄精组能改善CRF大鼠的贫血情况,降低血白细胞(P0.05),升高血红蛋白和红细胞(P0.01),可显著降低CRF大鼠血清AT-Ⅱ、ET(P0.01),升高CRF大鼠血清6-Keto-PGF1а。结论:制黄精可改善慢性肾衰竭大鼠肾血液动力,从而显著地改善肾功能。

Abstract:Objective:To observe the polygonatum effects on hemodynamics of rats with chronic renal failure,thus exploring the mechanism of protective effect on kidney of rats with diabetic nephropathy.Method:Take 21 clean level SD male rats and divide them into the sham operation group and the model group,through the 5/6 nephrectomy manufacturing method to make the chronic renal failure(CRF)animal model.Two weeks later,blood sample was taken from tail veins,serum creatinine determination(Scr)and urea nitrogen(BUN)values were tested.According to the Scr value,the model rats were randomly divided into pathological group,polygonatum group and the sham operation group.8 weeks after feeding or treatment,he rats were sacrificed,then hemoglobin(Hb),red blood cell(RBC),white blood cell(WBC),serum creatinine(Cr),blood urea nitrogen(BUN),24 hours urine protein quantity(Pro),endothelin(ET),calcitonin gene related peptide(CGRP),angiotensin I(AT-I),angiotensin II(AT-II),TXB2 and 6-Keto-PGF1 were determined.Results:The kidney function and the routine blood test result in polygonatum group were obviously improved.Compared with pathological group,polygonatum group could improve the anemia condition,reduce the white blood cells(P<0.05)and rise the Hb as well as red blood cells(P<0.01);levels of AT-Ⅱ and ET in serum could be obviously decreased and 6-Keto-PGF1а level be increased.Conclusion:The polygonatum can improve hemodynamics of kidney of chronic renal failure rats so as to significantly improve renal function.
