蜂贝化瘀胶囊对糖尿病大鼠肾脏细胞Fas和Fas-L表达的影响

目的:研究中药蜂贝化瘀胶囊对糖尿病大鼠肾脏细胞凋亡及Fas和Fas-L表达的影响,探讨该药对糖尿病肾病的防治机制.方法:大鼠腹腔注射链脲佐菌素复制糖尿病大鼠模型,用蜂贝化瘀胶囊对糖尿病大鼠进行干预治疗,采用全自动生化分析仪测定大鼠血、尿代谢指标,原位末端标记法检测肾脏细胞凋亡,免疫组化和流式细胞术检测肾皮质Fas和Fas-L的表达.结果:模型组血糖(Glu)、尿素氮(BUN)、尿蛋白(Upro)较正常组明显升高(P＜0.01),中药组则有明显改善.模型组较正常组大鼠肾小球、肾小管凋亡细胞数明显增多(P＜0.01),Fas和Fas-L的表达显著增强(P＜0.01);中药组较模型组大鼠凋亡细胞数明显减少(P＜0.01),Fas和Fas-L表达减弱(P＜0.01).结论:蜂贝化瘀胶囊可能通过影响凋亡相关蛋白Fas和Fas-L的表达而抑制肾脏细胞凋亡,从而发挥对肾脏的保护作用.

Effects of Fengbei Huayu Capsule on Expressions of Fas and Fas-L in Renal Cells of Diabetic Rat

Objective:To investigate effects of Fengbei Huayu Capsule on apoptosis of renal cells and expression of Fas and Fas-L in renal cells of diabetic rat,so as to study mechanism in prevention and treatment of diabetic nephropathy.Methods:Diabetic rat model was made by intraperitoneal injection of streptozotocin,and treated with Fengbei Huayu Capsule.Biochemical indexes of blood and urine were determined with full-automatic biochemical analyzer and apoptosis of renal cells was detected with in situ end labelling method.Expression of Fas and Fas-L in renal cortex was detected with immunohistoehemical method and flow cytometry. Results:Blood glucose(Glu),blood urea nitrogen(BUN)and urinary protein(Upro)of the model gronp increased significantly as compared with those of the normal group(P<0.01),and improved significantly as compared with Chinese medicine group(P<0. 01).Apoptosic cells of glomerulus and renal tubules in the model group increased significantly(P<0.01)and expression of Fas and Fas-L raised as compared with the normal group(P<0.01);The apoptosic cells in Chinese medicine group decreased significantly (P<0.01)and expression of Fas and Fas-L deceased significantly(P<0.01)as compared with the model group.Conclusion: Fengbei Huayu Capsule inhibits apoptosis of renal cells by means of influencing expression of apoptosis-related proteins Fas and Fas-L,so as to exert protective action on the kidney.
