小葱葱白提取物对人脐静脉内皮细胞一氧化氮及一氧化氮合酶的影响

摘    要:目的观察小葱葱白提取物对人脐静脉内皮细胞(HUVECs)分泌一氧化氮(NO)和内皮型一氧化氮合酶(eNOS)的影响,探讨其对HUVECs血管活性分子分泌的调节作用。方法从小葱葱白中提取活性成分,体外培养HUVECs用不同浓度(100g/L,50g/L,25g/L,12.5g/L)的葱白提取物分别作用于HUVECs。观察细胞形态,MTT法观察葱白提取物对HUVECs活性的影响,比色法测定各组的NO含量,RT-PCR法及Western Blot法测定内皮型eNOS的表达水平。结果和对照组比较,小葱葱白提取物对内皮细胞形态和活性无明显影响,能增加HUVECs释放NO及eNOS生成(P<0.05或P<0.01)。结论小葱葱白提取物可能通过增加eNOS生成而提高HUVECs释放NO,从而保护内皮功能。

Abstract:Objective To investigate the effect of fistular onion stalk extraction on nitrous oxides(NO)and nitrous oxides synthase(NOS)in cultured human umbilical vein endothelium cells(HUVECs).Methods The active composition was extracted from fistular onion stalk.The cultured HUVECs were incubated in different concentration extractions of fistular onion stalk in vitro(100 g/L,50 g/L,25 g/L,12.5 g/L),respectively.Cells morphology was observed by light microscope.Cells viability was assessed by MTT assay.NO content was measured by the colorimetry.The expression level of endothelial type NOS(eNOS)was assessed by RT-PCR and Western Blot analysis.Results Cells morphology and viability were not affected by the extraction of fistular onion stalk.The level of NO and the activity of eNOS increased by extraction of fistular onion stalk significantly(P<0.05 or P<0.01).Conclusion The extraction of fistular onion stalk could increase the level of NO by the enhancement the activity of eNOS.It might have the protection on endothelial function.
