钩藤碱对帕金森大鼠脑内SOD、DA、MDA表达的调节作用

摘    要:目的探讨钩藤碱在帕金森病(PD)大鼠模型中对纹状体多巴胺(DA)、超氧化物歧化酶(SOD)、丙二醛(MDA)水平表达的调节作用。方法首先从中药钩藤中分离提取出钩藤碱。其次采用脑内定位注射6-OHDA制备部分损伤PD大鼠模型,大鼠随机分为3组(各组10只大鼠),分别为正常对照组、模型对照组、钩藤碱组。用生化法对大鼠脑中DA及血清中SOD、MDA活性进行测定,分析钩藤碱对其活性的影响。结果给药30d后,钩藤碱组DA水平密度明显高于模型对照组(P〈0.01)。在SOD水平方面,钩藤碱组显著高于模型对照组(P〈0.05);而MDA含量,钩藤碱组与正常对照组接近,但显著低于模型对照组(P〈0.05)。结论钩藤碱对帕金森大鼠脑内SOD、DA、MDA表达的具有调节作用,提示其作为帕金森症的治疗或者预防药物具有一定的开发价值。

Abstract:Objective To study the regulation of rhynchophylline on the expression levels of dopamine(DA),superoxide dismutase(SOD)and malondialdehyde(MDA)in rats with Parkinson disease(PD).Methods First to extracted rhynchophylline from traditional Chinese medicine uncaria.The second part is to injury PD rat model by Brain positioning injection of 6-OHDA,compare and analyze normal group,model group and rhynchophylline group.Determine DA,SOD in serum and MDA activity in rats' brains by biochemical method,and analyze the impact of rhynchophylline on its activity.Results After 30days drug administration,DA density level of rhynchophylline group was significantly higher than that of model group(P〈0.01).At the SOD level,rhynchophylline group was significantly higher than that in model group(P〈0.05).Meanwhile the content of MDA compared with the normal group,rhynchophylline group and normal group ware close and significantly lower than that of model group(P〈0.05).Conclusion Rhynchophylline could adjust SOD,DA and MDA in brains in rats with Parkinson disease(PD).It shows a value of development in Parkinson disease treatment or prevention of drug.
