Abstract:AIM: To investigate the effect of simulated weightlessness on cell cycle and proliferation of human gastric carcinoma cell line SGC-7901. METHODS: A rotating clinostat was used to simulate weightlessness. The expression of PCNA was examined by immunohistochemical staining. The changes of cell cycle were examined by flow'cytometry. RESULTS: The expression of PCNA did not change significantly after 12 h, 24 h, and 36 h of simulated weightlessness, but decreased significantly after 48 h and 72 h (P<0.05 and P<0.01, respectively). The cell cycle of 12 h, 24 h, and 36 h groups did not change significantly. However, the cells in G0+G1 phase in 48 h and 72 h groups were increased significantly (P<0.01 and P<0.05, respectively), while the cells in S+G2+M phase were decreased significantly (P<0.01 and P<0.05). CONCLUSION: Simulated weightlessness by a clinostat can prevent the transition of cells from G1 phase to S phase, and thus the proliferation of SGC-7901 cells was inhibited.
